Treatment protocol 32D cells were transduced with MPL and Calr (WT or ins5). Cells were starved from IL3 over night in RPMI+FCS+P/S.
Growth protocol cells were cultured at 37°C, 5% CO2, in RPMI supplemented with 10% FCS, 1% P/S and 5ng/ml IL3
Extracted molecule total RNA
Extraction protocol Cell were washed with PBS to remove any IL3 and cultured over night. Cell were harvested from overnight culture and total RNA was isolated using QIAzol method.
Label Biotin
Label protocol We used the Affymetrix WT Pico kit for cDNA synthesis, amplification, cRNA synthesis, second-cycle cDNA synthesis, as well as fragmentation and labeling with Biotin according to manufacturer's instructions.
 
Hybridization protocol Labeled fragments were hybridized to Applied Biosystems Clariom S Mouse Arrays for 16h at 45°C with 60 rpm in an Affymetrix Hybridization Oven 645.
Scan protocol After washing and staining, the arrays were scanned with the Affymetrix GeneChip Scanner 3000 7G. CEL-files were produced from the raw data with Affymetrix GeneChip Command Console Software.
Description Gene expression from WT mouse CXCL12-abundant reticular cells
Data processing CEL files were analysed using the oligo R package. RMA method, log2 transformed were used to normalized the probe intensities.
