SKH-1 mouse skin (3 mice per treatment group) was treated topically (with 200µl) on their backs either with vehicle (acetone) or 8 mM BMS-202 [all 3 times: 24 hr pre, 1 hr pre, and immediately post acute SSL (solar simulated UV light) treatment] and harvested 24 hr after acute SSL. SSL irradiation of SKH-1 mice was performed at a dose of 90 kJ/m2 UVA/ 6 kJ/m2 UVB.

All SKH-1 mice were housed and treated in accordance with The University of Arizona Animal Care and Use Committee standards under an approved protocol (#08-153).

Total RNA was extracted from murine full tickness skin tissues using Qiagen RNeasy Mini kit according to manufacturer instructions.

All samples were run targeting 100 ng of total RNA (19 h at 65°C) prior to being processed on the nCouner FLEX Analysis System.

Samples were run on the nCounter Prep Station with high sensitivity settings followed by imaging on the Digital Analyzer with FOV (Field of View) data resolution set to 555.
