Extraction protocol |
Total RNA was isolated from cell lysates using the RNAzol RT protocol as per the manufacturer’s instructions RNA Illumina-ready libraries were generated using NEBNext Ultra II RNA Prep reagents (New England BioLabs). Using oligo dT beads (Invitrogen), poly-adenylated RNA was purified and subsequently fragmented, followed by double-stranded cDNA synthesis, end repair, and adapter ligation. The ligated DNA was then barcoded and amplified by a limited cycle PCR and the libraries were sequenced using a paired-end 150-base protocol on either a HiSeq 4000 or a NovaSeq 6000 (Illumina).
