Extraction protocol RNA was purified using RNACleanXP Solid Phase Reversible Immobilization (SPRI) beads (Beckman Coulter). Beads with bound RNA were re-suspended in 5 ul of RSB buffer.
Single-cell RNA sequencing was performed by Medgenome using a modified version of the ClonTech SMART-Seq Single Cell Kit that utilized miniaturization to ΒΌ volumes using a MANTIS Liquid Handler (Formulatrix). Amplified cDNA was fragmented and appended with dual-indexed barcodes using Illumina Nextera kits.

Data processing The fastq files were aligned to MacaM v7 using STAR aligner v 2.5.2b. The raw counts from ReadsPerGene output of STAR was used for downstream analysis using Seurat v4.0.4.

Supplementary files format and content: The ReadsPerGene output from STAR for each cell was combined as separate columns in ReadCounts.txt.The MacaM v7.8.2 gtf was used to convert gene id to gene names.
