Treatment protocol For the banding procedure, 10-week-old male mice were anesthetized with a mixture of ketamine (90 mg/kg) and xylazine (10 mg/kg). The aortic arch was carefully exposed, and a 7-0 silk suture was used to tightly constrict the aorta between the brachiocephalic trunk and the left carotid artery over a wire of 0.3mm in diameter is put on the aorta. It is banded with the aorta together and is removed. Sham-operated mice were subjected to the same procedure without aortic banding. The hearts were isolated 4 weeks after the operation. The heart weight/body weight (HW/BW) ratio and cross-sectional area of myocytes within the left ventricular wall accordantly showed a greater than 1.5-fold increase in WT TAC compared to WT sham. KO mice showed significantly less cardiac hypertrophy than WT TAC.
Growth protocol Standard breeding was performed until 4 weeks after the operation.

Extraction protocol Total RNA samples were extracted from the left ventricles using ISOGEN II reagent (Nippon Gene).

Label protocol Amino Allyl aRNA was synthesized using the Amino Allyl MessageAmp II aRNA Amplification Kit (Ambion #1753). Cye dye coupling and fragmentation were performed according to the protocol supplied by TORAY Industries, Inc.
 
Hybridization protocol Hybridized for 16 h at 37 C with rotary shaking (250 rpm). Hybridization buffer and washing protocol were according to the protocol supplied by TORAY Industries, Inc.
Scan protocol ScanArray Express (PerkinElmer Japan Co., Ltd.) was used for scanning. Images were quantified using GenePix Pro version 6.0 (Axon Instruments).

Data processing The raw data of each spot was normalized by substitution with the mean intensity of the background signal determined by all blank spots' signal intensities of 95% confidence intervals.
