Extraction protocol Bovine mammary gland were removed, flash frozen on dry ice, and RNA was harvested using Trizol reagent. Illumina TruSeq Small RNA Sample Prep Kit (Cat#FC-122-1001) was used with 1 ug of total RNA for the construction of sequencing libraries.

Data processing Small RNA reads were processed using Illumina’s Genome Analyzer and ACGT101-miR program was then used to clean the sequencing data.
the sequencing data were filtered out the sequencing data to mRNA, RFam, Repbase, and then mapped to miRbase. The mapped data were then aligned to genome and EST, databases revealing genome locations or annotations.
The rests of unmapped data were mapped to genome and EST, and secondary structure were predicted using UNAFold software
IDEG 6 was used to identify significant differentially expressed miRNAs
