Extraction protocol Total RNA from three independant cultures were extracted using the SV Total RNA isolation System (promega)
Label Cy-3
Label protocol Incorporation of cyanine-3 and cyanine-5 was performed during reverse transcription of total RNA using label star reverse transcriptase (quiagen). Labelled cDNA from the microarray experiment was purified on minElute spin columns (quiagen).
 
Hybridization protocol Hybridation was carried out in an automatic hybridization chamber ( Discovery from Ventana Medical System) for 10h at 42°C. After hybridization, slides were washed in 2xSSC/0,2%(v/v)SDS, immersed briefly in iso-propanol and then dried under a stream of air.
Scan protocol The hybridization signal were detected by scanning using GenePix 4000B laser Scanner (Axon Instruments) and transformed to numerical values using the integrated GenePix software version 3.01
Description WT harvested in exponential phase biological replicate 1
Data processing R computing environnement was used for background subtraction and weighted global median and quantile normalization was performed
