tRF & tiRNA sequencing of Aspergillus fumigatus

(Submitter supplied) Aspergillus fumigatus is an important human pathogen and a leading fungal killer. This study aimed to determine the tRNA fragment and tRNA half repertoire of A. fumigatus in wild-type conidia and mycelium grown for 24 or 48 hours in liquid culture.

The oxidative stress response highly depends on glucose and iron availability in Aspergillus fumigatus

(Submitter supplied) Pathogens have to cope with oxidative, iron- and carbon-limitation stresses in the human body. To understand how combined iron-carbon limitation alters oxidative stress response, Aspergillus fumigatus was cultured in glucose-peptone or peptone containing media supplemented or not with deferiprone as iron chelator. Changes in the transcriptome in these cultures were recorded after H2O2 treatment. Responses to oxidative stress were highly dependent on the availability of glucose and iron. more...

Fludioxonil-induced differential gene expression in Aspergillus fumigatus and its DtcsC and Dskn7 mutants

(Submitter supplied) The antifungal compound fludioxonil targets the fungal specific group III hybrid histidin kinaseTcsC of A. fumigatus, which in turn activate the High Osmolarity Glycerol (HOG) pathway and leads to drastic swelling of the fungal cells. We investigated the different responses of the AfS35 (wild type), the completely resistant mutant strain DtcsC and the partial resistant mutant strain Dskn7 to fludioxonil.

Quantification of Bir1-dependent differential gene expression in swollen Aspergillus fumigatus conidia

(Submitter supplied) This study aimed to understand the differences in gene expression when comparing Aspergillus fumigatus swollen conidia that overexpress an artifical Bir1 construct (strain published in Shlezinger et. al 2017) versus a parental control strain in RPMI + 0.5ug/mL voriconazole. The purpose of this experiment was to understand what basal expression differences may drive a decreased susceptibility to hydrogen peroxide upon subsequent treatment (not tested in this submission). more...

Stress responses elicited by glucose withdrawal in Aspergillus fumigatus

(Submitter supplied) Glucose is a widely used carbon source in laboratory practice to culture Aspergillus fumigatus, however, glucose availability is often low in its “natural habitats” including the human body. We used a physiological–transcriptomical approach to reveal differences between A. fumigatus Af293 cultures incubated on glucose, glucose and peptone, peptone (carbon limitation), or without any carbon source (carbon starvation). more...

Dual-RNA sequencing of dendritic cells challenged with Aspergillus fumigatus

(Submitter supplied) Dendritic cells and A. fumigatus were co-cultured with two different MOIs (1 or 0,5) for 2, 4 or 6 hours. Single-cultures of dendritic cells and A. fumigatus serve as controls. RNA was isolated and processed for next generation sequencing.

The fungal CCAAT-binding complex and HapX display highly variable but evolutionary conserved synergetic promoter-specific DNA recognition

(Submitter supplied) To sustain iron homeostasis, microorganisms have evolved fine-tuned mechanisms for uptake, storage and detoxification of the essential metal iron. In the human pathogen Aspergillus fumigatus, the fungal-specific bZIP-type transcription factor HapX coordinates adaption to both iron starvation and iron excess and is thereby crucial for virulence. Previous studies indicated that a HapX homodimer interacts with the CCAAT-binding complex (CBC) to cooperatively bind bipartite DNA motifs; however, the significance of the HapX and CBC interaction and the mode of HapX-DNA recognition had not been resolved. more...
