Discovery of genes associated with whirling disease infection and resistance in rainbow trout using expression profiling

(Submitter supplied) Whirling disease, caused by the pathogen Myxobolus cerebralis, afflicts several salmonid species. Rainbow trout are particularly susceptible and suffer high mortality rates. The disease is persistent and spreading in hatcheries and natural waters of several countries, including the U.S., and the economic losses associated with whirling disease have been extremely large. In this study, gene expression profiling was conducted for resistant and susceptible rainbow trout strains following pathogen exposure with the primary objective of identifying specific genes associated with whirling disease resistance. more...

Cell line stimulated with recombinant cytokines interleukin-1b and interferon gamma.

(Submitter supplied) Interleukin 1b and interferon g are two cytokines that are central to the inflammatory response. They both have a broad range of activity, each directing the immune response to be towards either the innate response (IL-1b and IFN-g) or the cellular response (IFN-g), however invivo there is a lot of subtle interactions. Microarray analysis has been used to characterize the transcriptional response to recombinant rainbow trout IL-1b and IFN-g in a rainbow trout macrophage cell line RTS-11. more...

Salmonid host response to Hematopoietic Necrosis virus: cellular receptors, viral control and novel pathways of defence

(Submitter supplied) The transcriptional response of Atlantic salmon (Salmo salar) to infectious hematopoietic necrosis (IHN) virus was elucidated using 16,008 gene GRASP cDNA microarrays. S. salar were exposed to the IHN virus in a waterborne challenge, and kidney samples from five fish sampled on each of days 0, 1, 5 and 9 were analysed in the microarray study. Validation of nine of the significant genes was conducted on fish sampled on alternate days from 1 to 13 and compared to the unchallenged control sampled on day 0. more...

Microarray gene expression analysis in atrophying rainbow trout muscle: An unique non-mammalian muscle degradation model

(Submitter supplied) Muscle atrophy is a physiological response to diverse physiological and pathological conditions that trigger muscle deterioration through distinct cellular stimuli. Despite different physiological signals, the resulting biochemical changes in atrophying muscle share many common cascades. Muscle deterioration as a physiological response to the energetic demands of fish spawning represents a unique model for studying the mechanisms of muscle degradation in non-mammalian animals. more...

The effects of chronic immune stimulation on muscle growth in rainbow trout

(Submitter supplied) Successful production of aquaculture species depends on efficient growth with low susceptibility to disease. Therefore, selection programs have focused on rapid growth combined with disease resistance. However, chronic immune stimulation diminishes muscle growth (a syndrome referred to as cachexia), and decreases growth efficiency in production animals, including rainbow trout. In mammals, recent results show that increased levels of pro-inflammatory cytokines, such as those seen during an immune assault, specifically target myosin and MyoD and inhibit muscle growth. more...

Gene Expression Profiling Following DNA Vaccination of Rainbow Trout against Infectious Hematopoietic Necrosis Virus

(Submitter supplied) The Infectious Hematopoietic Necrosic Virus (IHNV) DNA vaccine is based on the viral glycoprotein gene (G gene) and induces a non-specific anti-viral immune response and long-term specific immunity against IHNV in salmonid fishes. This study characterized gene expression responses associated with the early anti-viral response. Homozygous rainbow trout were injected intra-muscularly (I.M.) with vector DNA or the IHNV DNA vaccine. more...
