The effect of mug174 deletion or red1 deletion mutation on gene expression in the fission yeast Schizosaccharomyces pombe

(Submitter supplied) Our characterization of Mug174 using genetic and biochemical approaches revealed that Mug174 is the Coilin (an integral component of the Cajal body in multicellular organisms) ortholog in the fission yeast S. pombe. To investigate the Mug174 role in shaping transcriptome, we delete the mug174 gene and performed RNA-seq. The Rtf1/Prf1-dependent histone modification axis and Rpb1 C-terminal domain phosphorylation counteract multi-drug resistance in fission yeast [RNA-seq]

(Submitter supplied) RNA polymerase II (RNAPII) transcription elongation directs an intricate pattern of histone modifications. Formation of this pattern involves a highly conserved regulatory axis, key components of which are the elongation factor Rtf1, monoubiquitylation of histone H2B (H2Bub1), and methylation of histone H3 on lysine 4 (H3K4me). Direct contact between Rtf1 and the ubiquitylation complex Rad6-Bre1 stimulates H2Bub1, which in turn promotes H3K4me by stimulating the activity of the Set1C/COMPASS methyltransferase complex. more...

The chromatin remodeler Ino80 mediates alternative RNAPII pausing site determination

(Submitter supplied) Promoter-proximal pausing of RNAPII is a critical step in early transcription elongation to precisely regulate gene expression. Here, we observed evidence of promoter-proximal pausing like distributions of RNAPII in S. cerevisiae. We find that Ino80p loss caused the transition of RNAPII pausing to the alternative pausing site at which is tightly associated with the +1 nucleosome. These Ino80p dependent genes showed better nucleosome positioning around the main pausing site which suggested the regulation of RNAPII pausing by Ino80p in a nucleosome context-dependent manner. more...

Investigation of alteration in gene expression during oxidative stress in ∆wee1S. pombe cells

(Submitter supplied) Treating wild-type S. pombe cells and ∆wee1 mutant with 20mM H2O2, we observed that aberrant Cdc2 activity, as in case of ∆wee1 cells can lead to major changes in the Spc1 dependent global transcriptional response to oxidative stress.
