Dynamic changes in gene expression in vivo predict prognosis of tamoxifen-treated patients with breast cancer

(Submitter supplied) Tamoxifen is the most widely prescribed anti-estrogen treatment for patients with ER-positive breast cancer. However, there is still a need for biomarkers that reliably predict endocrine sensitivity in breast cancers and these may well be expressed in a dynamic manner. In this study we assessed gene expression changes at multiple time points (days 1, 2, 4, 7, 14) after tamoxifen treatment in the ER-positive ZR-75-1 xenograft model that displays significant changes in apoptosis, proliferation and angiogenesis within 2 days of therapy. more...

SREBP1c and mock virus expression profiles in AG01518 human fibroblasts

(Submitter supplied) Sterol-regulatory element binding proteins (SREBP) control the expression of most genes involved in fatty acid and cholesterol metabolism. In this report, using microarrays, we showed that SREBP1 also induced genes unrelated to lipid biosynthesis, such as heme oxygenase 1 (HMOX1), the phosphatidylinositol-3 kinase regulatory subunit p55-gamma, synaptic vesicle glycoprotein 2A (SV2A) and COTE1 (C1orf2). more...

PDGF stimulation kinetics

(Submitter supplied) Subconfluent AG01518 fibroblasts were starved for 16 hours in the presence of 0.1% BSA, and then stimulated for 1, 4, 10 or 24 hours with PDGF-BB (10 ng/ml in starvation medium), or left untreated for the same periods of time. Total RNA was isolated using the RNeasy kit (Qiagen). The RNA quality was checked by formaldehyde agarose gel electrophoresis (RNeasy protocol, Qiagen). Total RNA (40 mg) from cells treated for a given period of time with PDGF or starvation medium alone were labelled in reverse transcription reactions (Superscript II kit, Invitrogen) with dCTP-Cy5 and dCTP-Cy3, respectively (Amersham). more...

The effect of Ras on the signalling networks in senescence

(Submitter supplied) Activated Ha-ras was introduced to HMF3A cells. HMF3A cells are conditionally immortalised human mammary fibroblasts. They undergo co-ordinated senescence upon inactivation of LT antigen, when cultured at 39C. The introduction of Ras did not prevent this but affected the molecular profile. HMF3A_RNAi

(Submitter supplied) HMF3A cells, created from adult human mammary fibroblasts by immortalisation with a thermolabile SV40 large T antigen and the catalytic sub-unit of human telomerase, undergo co-ordinated induction of cellular senescence upon inactivation of T antigen. The pSUPER-retro vector system (Brummelkamp, 2002) was used to selectively target genes for mRNA degradation in an attempt to determine if they had a role, in the changes to the transcriptome upon LT inactivation. more...

GSEp53_v_Con

(Submitter supplied) HMF3A cells, created from adult human mammary fibroblasts by immortalisation with a thermolabile SV40 large T antigen and the catalytic sub-unit of human telomerase, undergo co-ordinated induction of cellular senescence upon inactivation of T antigen. Given the importance of p53, we sought to determine what gene changes were affected if its activation was inhibited. To do this we used the Genetic Suppressor Element (GSE)56 which inhibits the transcriptional activity of p53 (Ossovskaya, 1996). more...

HMF3A_with_3Dwt

(Submitter supplied) HMF3A cells, created from adult human mammary fibroblasts by immortalisation with a thermolabile SV40 large T antigen and the catalytic sub-unit of human telomerase, undergo co-ordinated induction of cellular senescence upon inactivation of T antigen. HMF3A cells cease proliferating by 4 days at 39°C. We directly compared the gene expression profiles of cells at 33.5°C and after shift up to 39.5°C for 7 days (10 arrays, 3 biological replicates). more...

Prim_Fibs

(Submitter supplied) To determine changes in gene expression in primary fibroblasts undergoing replicative senescence, a direct comparison between early passage proliferating cells and senescent cells was performed. Frozen stocks of the primary fibroblasts (HMF3) from which the lines HMF3A and HMF3Dwt were derived (O'Hare, PNAS:98, 2001) were serially passaged twice through to senescence to yield 2 independent sets of RNA. more...
