Inferring Epitopes of a Polymorphic Antigen Amidst Broadly Cross-Reactive Antibodies Using Protein Microarrays: a Study of OspC Proteins of Borrelia burgdorferi

(Submitter supplied) Epitope mapping studies aim to identify the binding sites of antibody-antigen interactions to enhance the development of vaccines, diagnostics and immunotherapeutic compounds. However, mapping is a laborious process employing time- and resource-consuming ‘wet bench’ techniques or epitope prediction software that are still in their infancy. For polymorphic antigens, another challenge is characterizing cross-reactivity between epitopes, teasing out distinctions between broadly cross-reactive responses, limited cross-reactions among variants and the truly type-specific responses. more...

OspC Protein Microarray

(Submitter supplied) Primers used for amplifying full-length ospC alleles had sequences corresponding to positions 16903 through 16925 (Forward) and 17529 through 17558 (Reverse) of cp26 of the B31 strain. For amplification of subcloning targets for protein expression the Forward primer eliminated the signal peptide and 4 different Reverse primers (to accommodate 3’ end nucleotide polymorphisms amongst ospC alleles), corresponding to nucleotide positions 614 through 633 were used. more...
