Various prostate cell types

Analysis of basal, luminal secretory, stromal fibromuscular, and endothelial cells of the prostate. Cell types separated with magnetic cell sorting (MACS) based on cell-type specific cluster designation (CD) antigens. Results identify gene expression signatures at the level of individual cell types.

Prostate cell specific expression

(Submitter supplied) Luminal, basal, stromal, and endothelial cells were MACS sorted from whole tissue. Targets from five biological replicates of each were generated and the expression profiles were determined using Affymetrix U133 Plus 2.0 arrays. These data represent cell specific transcriptomes. Distinctive gene expression of prostatic stromal cells cultured from diseased versus normal tissues

(Submitter supplied) To obtain a comprehensive view of the transcriptional programs in prostatic stromal cells of different histological/pathological origin, we profiled 18 adult human stromal cell cultures from normal transition zone (TZ), normal peripheral zone (PZ), benign prostatic hyperplasia (BPH), and prostate cancer (CA) using cDNA microarrays. Set of arrays organized by shared biological context, such as organism, tumors types, processes, etc. more...

TGFβ Signaling in Prostate Stroma Supports Carcinoma Growth by Upregulating Stromal Genes Related to Tissue Remodeling

(Submitter supplied) Increasing evidence points to an active stromal involvement in cancer initiation and progression. Cytokines derived from tumor cells are believed to modulate stromal cells to produce growth and angiogenic factors which in turn provide the tumor with the necessary microenvironment for expansion and invasion. Transforming growth factor beta (TGFβ) has been implicated as a candidate cytokine to mediate this communication. more...

Expression profile of early and mid-secretory human endometrium revealed by RNA-seq: a paired-sample cohort study

(Submitter supplied) Inner uterine lining or endometrium is a unique constantly self-renewed adult tissue that is vital for embryo implantation. As the footrace for universal endometrial receptivity markers continues, RNA-seq remains the most powerful tool for transcriptomic marker discovery. In this study, we aimed to elucidate endometrial maturation mechanisms at transcriptomic level and identify novel robust biomarker candidates for endometrial receptivity in a multicentre study. more...

Gene profiling of primary cultures from human prostate tumors

(Submitter supplied) The histopathological and molecular heterogeneity of prostate cancer and the limited availability of human tumor tissue make unraveling the mechanisms of prostate carcinogenesis a challenging task. Our goal was to develop an ex vivo model that could be reliably utilized to define a prognostic signature based on gene expression profiling of cell cultures that maintained the tumor phenotype. To this end, we derived epithelial cultures from tissue explanted from 59 patients undergoing radical prostatectomy or cistoprostatectomy because of Prostate Benign Hyperplasia/Prostate Cancer or Bladder Carcinoma. more...

Transcriptomes of human bladder cells and cells in bladder cancer

(Submitter supplied) Characterization of the gene expression profiles of specific cell populations of the human urinary bladder provides an important set of research tools for the study of cellular differentiation and the cancer process. The transcriptome is a definitive identifier of each individual cell types. Surgically resected tissue was digested by collagenase and the different cell types were sorted by antibodies to cluster designation (CD) cell surface antigens. more...

Transcriptome of the bone metastasis associated stroma

(Submitter supplied) The reciprocal interaction between cancer cells and the tissue-specific stroma is critical for primary and metastatic tumor growth progression. Prostate cancer cells colonize preferentially bone (osteotropism), where they alter the physiological balance between osteoblast-mediated bone formation and osteoclast-mediated bone resorption, and elicit prevalently an osteoblastic response (osteoinduction). more...

Differential Gene Expression on Human LNCaP Cells Following Treatment with DHEA versus DHT

(Submitter supplied) ABSTRACT The effects of DHEA were compared with those of DHT using gene expression array profiles in human LNCaP prostate cancer cells. LNCaP cells were exposed to DHEA (300 nM), DHT (300 nM), or vehicle for 48 hrs, and mRNA was isolated. mRNA expression was measured using Affymetrix HU-95 gene chips in 3 experiments performed on different dates. Gene expression values for the two treatment groups and control were sorted in ascending order on the p-values corresponding to a variance stabilized Hotelling test, which measured the extent of differential RNA expression between control and either hormone treatment. more...

Single-cell transcriptome profiling of an adult human cell atlas of 15 major organs

(Submitter supplied) Background: As core units of organ tissues, cells of various types play their harmonious rhythms to maintain the homeostasis of the human body. It is essential to identify the characteristics of cells in human organs and their regulatory networks for understanding the biological mechanisms related to health and disease. However, a systematic and comprehensive single-cell transcriptional profile across multiple organs of a normal human adult is missing. more...

Recapitulation of Prostate Tissue Cell Type-specific Transcriptomes by an in vivo Primary Prostate Tissue Xenograft Model

(Submitter supplied) Purpose: Validate the cell type specific (Epithelial, Endothelial, and Stromal) transcriptome profile between prostate primary tissue xenografts and of fresh prostate tissue. Nude-Fox1nu mice were castrated and silastic tubing containing dry testosterone (T) was inserted providing “humanized” levels of circulating T. 5-8 pieces of fresh human prostate tissue was then inserted subcutaneously along the flank of the animal. more...
