Shp2 Signaling Suppresses Senescence in PyMT-induced Mammary Gland Cancer in Mice [Mouse430_2 array]

(Submitter supplied) In this study, we have used techniques from cell biology, biochemistry, and genetics to investigate the role of the tyrosine phosphatase Shp2 in tumor cells of MMTV-PyMT mouse mammary glands. Genetic ablation or pharmacological inhibition of Shp2 induces senescence, as determined by the activation of senescence-associated β-gal (SA-β-gal), cyclin-dependent kinase inhibitor 1B (p27), p53, and histone 3 trimethylated lysine 9 (H3K9me3). more...

Mouse tumors: Chaos3 Mammary Tumors (MTs) vs. Controls

(Submitter supplied) CNV profiling of tumors obtained from our Chaos3 mouse model for spontaneous breast cancer. The goal of this experiment was to determine copy number variations that were specific to MTs derived from this mouse model, when comapared to non-MTs.

Next Generation Sequencing of 23116 MT (low Arid1a expression) vs AB-C1 and AB-C2 (high Arid1a expression) Transcriptomes

(Submitter supplied) The goal of this study was to identify important genetic pathways that are altered in mammary tumor cells upon over-expression of the tumor suppressor gene Arid1a. The results of this experiment revealed that Arid1a helps regulate key cell-cycle checkpoint and growth regulatory pathways, either directly or indirectly. This helped explain in part the significant decrease in cell proliferation and tumor growth phenotypes observed both in vitro and in vivo, when comparing the same samples analyzed here by RNA-seq (untransduced replicates vs. more...

Genome wide expression analysis of FACS-isolated Shp2 mutant neonatal myogenic progenitor cells

(Submitter supplied) The equilibrium between proliferation and quiescence of myogenic progenitor and stem cells is tightly regulated to ensure appropriate muscle growth and repair. The non-receptor tyrosine phosphatase Shp2 (Ptpn11) is an important transducer of growth factor and cytokine signaling. Here we combined complex genetic analyses, biochemical studies and pharmacological interference to demonstrate a central role of Shp2 in postnatal myogenesis of mice. more...

SUM159 cells grown in 2D cultures in the presence or absence of SHP2

(Submitter supplied) The first bona fide PTP proto-oncogene was the Src-homology 2 domain-containing phosphatase SHP2 (encoded by PTPN11), an ubiquitously expressed PTP that transduces mitogenic, pro-survival, cell fate and/or pro-migratory signals from numerous growth factor-, cytokine- and extracellular matrix receptors. In malignancies, SHP2 is hyperactivated either downstream of oncoproteins or by mutations. We provide analysis of the human mesenchymal triple-negative breast cancer cell line SUM159 and grown in 2D cultures for 15 days in the presence or absence of SHP2.
