ACLY and ACC1 Regulate Hypoxia-Induced Apoptosis by Modulating ETV4 via α-ketoglutarate

(Submitter supplied) In order to propagate a solid tumor, cancer cells must adapt to and survive under various tumor microenvironment (TME) stresses, such as hypoxia or lactic acidosis. To systematically identify genes that modulate cancer cell survival under stresses, we performed genome-wide shRNA screens under hypoxia or lactic acidosis. We discovered that genetic depletion of acetyl-CoA carboxylase (ACACA or ACC1) or ATP citrate lyase (ACLY) protected cancer cells from hypoxia-induced apoptosis. more...

Gene expression profile at single cell level of hematopoietic stem and progenitor cells (HSPCs) from mouse bone marrow cultured in M3434 StemCell methylcellulose media

(Submitter supplied) Hematopoiesis generates a variety of cell types after M3434 culture. We treated HSPCs with small-molecule inhibitors of Acly (SB204990), Gls (CB-839), or Vehicle (DMSO) and compared them to freshly isolated HSPCs by single-cell RNA-seq and single-cell ATAC-seq to examine the diverse cell types emerging from these conditions.

Effect of ACLY inhibition on LPS-induced EC gene profiles

(Submitter supplied) To investigate the regulatory role of ACLY activation in EC functions, we treated HUVEC with ACLY inhibitor BMS-303141 before LPS stimuli. We then performed gene expression profiling analysis using data obtained from RNA-seq of HUVEC lysates.

shRNA-mediated knockdown of ETV4 and MED25 in the prostate cell line PC3 reveals set of genes potentially coregulated by MED25 and ETV4

(Submitter supplied) Our in vitro binding studies support a model whereby MED25 exhibits multivalent interactions with a subset of related ETS factors, ETV1/4/5. We hypothesize that the interaction would allow for coregulation of genes by ETV1/4/5 and MED25, acting perhaps to link the ETVs to the Mediator complex. To explore this possibility, we compared the genome occupancy for FLAG-tagged MED25 and ETV4 in the prostate cancer cell line PC3, which overexpresses ETV4. more...

Genome-wide map of Acetyl-H3K27 marked chromatin in pancreatic cancer cells treated with vehicle and acetate under low pH condition

(Submitter supplied) We report the application of ChIP sequencing technology for high-throughput profiling of histone modification in pancreatic cancer cells. We obtained 49.7 million reads from acetylated-H3K27 modified chromatin immunoprecipitated DNA from vehicle and acetate-treated pancreatic cancer cells. We find that lysine 27 acetylation effectively discriminates genes that are expressed, or stably induced upon acetate treatment. more...
