Single-cell RNA-seq of Drosophila melanogaster male late third instar larval gonads

(Submitter supplied) The male larval gonad is a complex tissue made up of multiple cell types including germ cells and somatic cells. It is also a key developmental stage with the germline undergoing spermatogenesis. The diversity of cell types and transcriptional regulation of the developing gonad has only been broadly described. In order to get single cell resolution, we profiled late third instar larval testes of a reference Drosophila melanogaster genotype, w[1118] using single-cell RNA-Seq (scRNA-Seq). more...

Drosophila melanogaster late third instar larval ovary and gonadal fatbody polyA+ RNA-Seq profiling.

(Submitter supplied) Here we perform transcriptional profiling of late third instar larval ovaries and gonadal fatbody from Drosophila melanogaster w1118 flies. We designed this experiment to investigate the effect of protease treatment on dissected ovaries. We profiled ovaries without protease treatment with some fatbody still attached, fatbody alone dissected from around the ovaries, ovaries cleaned of fatbody using a papain/collagenase cocktail, and ovaries dissociated using a papain/collagenase cocktail. more...

Drosophila melanogaster late third instar larval testes and gonadal fatbody RNA-seq profiling

(Submitter supplied) In order to study the effect of protease treatment on dissected testes, we performed different strategies of cleaning and dissociation on Drosophila melanogaster w1118 larval testes. We collected testes without protease treatment with fatbody just attached around the gonad, fatbody alone dissected from around the testes, testes without fatbody and testes dissociated by either by Papain or Trypsin and Collagenase cocktail. more...

The ubiquitin-conjugating enzyme HR6B is required for maintenance of X chromosome silencing in mouse spermatocytes and spermatids

(Submitter supplied) Background: The ubiquitin-conjugating enzyme HR6B is required for spermatogenesis in mouse. Loss of HR6B results in aberrant histone modification patterns on the trancriptionally silenced X and Y chromosomes (XY body) and on centromeric chromatin in meiotic prophase. We studied the relationship between these chromatin modifications and their effects on global gene expression patterns, in spermatocytes and spermatids. more...

Drosophila melanogaster spermatogenesis expression profile: mitotic, meiotic and post-meiotic cells

(Submitter supplied) We conducted a genome-wide expression analysis of wild-type males using three cell populations isolated from mitotic, meiotic and post-meiotic phases of spermatogenesis in Drosophila melanogaster. Our approach was to directly isolate testis regions enriched with RNAs from each of the three specific germline phases. We used microarrays to detail the global gene expression profile in spermatogenesis and identified up- and down-regulated genes between two different spermatogenic phases in pairwise comparisons

A Pandas complex adapted for piRNA-guided transposon silencing and heterochromatin formation

(Submitter supplied) The repression of transposons by the Piwi-interacting RNA (piRNA) pathway is essential to protect animal germ cells. In Drosophila ovaries, Panoramix (Panx) enforces transcriptional silencing by binding to the target-engaged Piwi-piRNA complex, although the precise mechanisms by which this occur remain elusive. Here, we show that Panx functions together with a germline specific paralogue of a nuclear export factor, dNxf2, and its cofactor dNxt1 (p15) as a ternary complex to suppress transposon expression. more...

Direct interrogation of the role of H3K9 in metazoan heterochromatin function

(Submitter supplied) We replaced the endogenous histones of Drosophila melanogaster with histones containing an H3K9R mutation to interrogate the role of H3K9 in heterochromatin formation and function. We queried heterochromatin formation through Formaldhyde Assisted Isolation of Regulatory Elements coupled with sequencing to examine nucleosome occupancy and Heterochromatin Protein 1a ChIP sequencing to determine the localization of the major reader of H3K9me. more...

Piwi is required during Drosophila embryogenesis to license dual-strand piRNA clusters for transposon repression in adult ovaries [smallRNA-seq]

(Submitter supplied) Most piRNAs in the Drosophila female germline are transcribed from heterochromatic regions called dual-strand piRNA clusters. Histone 3 lysine 9 trimethylation (H3K9me3) is required for licensing piRNA production by these clusters. However, it is unclear when and how they acquire this permissive heterochromatic state. Although it has been suggested that piRNA cluster licensing is Piwi-independent, here we show that transient Piwi depletion in Drosophila embryos, using a refined knock-down system, results in H3K9me3 decrease at piRNA clusters. more...

Chromosome-specific maturation of the epigenome in the Drosophila male germline

(Submitter supplied) Spermatogenesis in the Drosophila male germline proceeds through a unique transcriptional program controlled both by germline-specific transcription factors and by testis-specific versions of core transcriptional machinery. This program includes the activation of genes on the heterochromatic Y chromosome, and reduced transcription from the X chromosome, but how expression from these sex chromosomes is regulated has not been defined. more...
