Next Generation Sequencing Facilitates Quantitative Analysis of yeast cells lacking either M-2 dsRNA or both L-A-lus and M-2 dsRNA viruses

(Submitter supplied) Here we determine the impact upon the loss of M-2 dsRNA alone and both L-A-lus along with M-2 on the host S. cerevisiae transcriptional changes. By performing profiling of the whole transcriptomes, we determined 486 genes differentially expressed after curing of yeast cells from M-2 dsRNA and 715 genes affected by the elimination of both M-2 and L-A-lus dsRNAs.

Elimination of Saccharomyces paradoxus viral dsRNA leads to transcriptional and phenotypic alterations

(Submitter supplied) Here we determine the impact upon the loss of M66 dsRNA alone and both L-A-66 along with M66 on the host S. paradoxus transcriptional changes. In Pet [L+M-] and [L-M-] cells we have found 941 and 651 differently expressed genes (DEGs), respectively. In total, there were 1234 and 535 DEGs in Spo [L+M-] and [L-M-] cells, respectively.

Small RNAs in S. cerevisiae reconstituted with RNAi

(Submitter supplied) The RNA interference (RNAi) pathway is found in most eukaryotic lineages but curiously is absent in others, including that of Saccharomyces cerevisiae. Here, we show that reconstituting RNAi in S. cerevisiae causes loss of a beneficial dsRNA virus, known as killer virus. Incompatibility between RNAi and killer viruses extends to other fungal species, in that RNAi is absent in all species known to possess dsRNA killer viruses, whereas killer viruses are absent in closely related species that retained RNAi. more...

Transcriptomic Analysis of Nonylphenol Effect on Saccharomyces cerevisiae

(Submitter supplied) Nonylphenol (NP), is a bioaccumulative environmental estrogen that is widely used as a nonionic surfactant. We have previously examined short-term effects of NP on yeast cells using microarray technology. In the present study, we investigated the long-term impacts of NP on Saccharomyces cerevisiae BY4742 cells by analyzing genome-wide transcriptional profiles using RNA-sequencing. We used 2 mg/L NP concentration exposure for 40 days. more...
