c-Maf-dependent Treg cell control of intestinal TH17 cells and IgA establishes host–microbiota homeostasis

(Submitter supplied) Foxp3+ regulatory T (Treg) cells are essential for immunological tolerance and homeostasis. In peripheral tissues, Treg cells acquire enhanced suppressive functions and co-opt distinct transcriptional modules, allowing context and tissue-dependent immune regulation. Here we show that the transcription factor c-Maf was highly expressed by effector Treg cells and controlled their IL-10 production. In the intestine, c-Maf was required for the differentiation of RORgt+ microbiota-dependent Treg cells, and restricted their production of inflammatory cytokines. more...

c-Maf-dependent regulatory T cells mediate immunological tolerance to a gut pathobiont

(Submitter supplied) Both microbial and host genetic factors contribute to the pathogenesis of autoimmune disease. Accumulating evidence suggests that microbial species that potentiate chronic inflammation, as in inflammatory bowel disease (IBD), often also colonize healthy individuals. These microbes, including the Helicobacter species, have the propensity to induce pathogenic T cells and are collectively referred to as pathobionts. more...

Loss of Bcl3 influences gene expression in Tregs and Treg subsets [Bcl3-WT_Treg_bulkRNAseq]

(Submitter supplied) Peripherally induced regulatory T cells (pTregs) expressing the retinoic acid receptor-related orphan-receptor gamma t (RORγt) are indispensable for intestinal immune homeostasis. Previous studies have determined the role of the atypical NFκB inhibitor Bcl3 in the development of colitis. In this study we analyzed the influence of Bcl3 on pTreg development and functionality in healthy mice and under colitogenic conditions. more...

Gene expression of Glut1 transgenic and control iTreg

(Submitter supplied) Effector (Teff) and regulatory (Treg) CD4 T cells undergo metabolic reprogramming to support proliferation and immune function. While Phosphatidylinositide 3-kinase (PI3K)/Akt/mTORC1 signaling induces the glucose transporter Glut1 and aerobic glycolysis for Teff proliferation and inflammatory function, mechanisms that regulate Treg metabolism and function remain unclear. We show that TLR signals that promote Treg proliferation increase Glut1, PI3K/Akt/mTORC1 signaling, and glycolysis. more...

Generation of RORγt+ antigen-specific T regulatory 17 (Tr17) cells from Foxp3+ precursors in autoimmunity

(Submitter supplied) Th17 cells are potent mediators in autoimmune diseases and RORγt is required for their development. Recent studies have shown that RORγt+ Treg cells in the gut regulate intestinal inflammation by inhibiting effector T cell function. In the current study, we report that RORγt+ Treg cells were also found in lymph nodes following immunization. Not only distinct from intestinal RORγt+ Treg in their transcriptomes, peripheral RORγt+ Treg cells were derived from Foxp3+ thymic Treg cells, in an antigen-specific manner. more...

c-Maf is an essential commitment factor for IL-17-producing gd T cells [ATAC-seq]

(Submitter supplied) IL-17-producing gd T (Tgd17) cells are early mediators of immunity and autoimmunity that undergo effector programming in the thymus. While Vg subset-restricted regulators of Tgd17 specialization are known, a universal type 17 commitment factor for all gd T cells remains undefined. In this study, we identify the transcription factor c-Maf as an essential and uniform regulator of Tgd17 differentiation and maintenance. more...

c-Maf-dependent transcripts in effector Tregs from Peyer's patches

An activation-induced immunoregulatory and tissue-residency program modulated by c-MAF in human memory Th17 cells [ChIP-seq] [RNA-seq]

(Submitter supplied) T cell effector function is largely mediated through the release of pro-inflammatory cytokines that need to be tightly regulated to avoid tissue damage. Here we show that the anti-inflammatory cytokine IL-10 is produced by a subset of recently-activated human Th17 cells and is controlled by the selective up-regulation of the transcription factor c-MAF. In addition, recently-activated IL-10+ Th17 cells upregulate immunoregulatory genes and genes characteristic of tissue resident T cells. more...
