Total RNA sequencing of KIR+ and KIR- NK cells from patients experiencing or not a Cytomegalovirus reactivation after haploidentical hematopoietic stem cell transplantation

(Submitter supplied) AIM: To investigate the adaptive properties of NK cells, by comparing the expression profiles of FACS-sorted KIR+ (CD158b1b2j) and KIR- NK cells from patients experiencing or not a Cytomegalovirus (HCMV) reactivation after haploidentical hematopoietic stem cell transplantation (h-HSCT). Comparison of Expression profile between CD3-CD56dimCD16+ CD57+ and CD57neg NK cells

(Submitter supplied) Thirty to 60% of CD56dimCD16bright NK cells in healthy adults express CD57, which is not expressed on immature CD56bright NK cells or fetal and newborn NK cells. Gene expression analysis for CD56- T, NK, CD56+ T cells, and iNKT cells

(Submitter supplied) A small subset of T cells also expresses kiler-cell immunoglobulin-like receptors (KIRs). We find that KIR+ T cells primarily reside in the CD56+ T population. However, little is known on how these cells are different from the conventional CD56- T, NK, and iNKT cells. We used microarray profiling to compare and determine the distinctive differences of CD56+ T cell and its KIR subsets when compared to the conventional CD56- T, NK and iNKT cells.

Malaria-driven expansion of adaptive-like functional CD56-negative NK cells correlates with clinical immunity to malaria

Expansion, persistence and efficacy of donor memory-like NK cells for the treatment of post-transplant relapse

(Submitter supplied) Relapse of AML occurs in up to 50% of patients after allogeneic hematopoietic stem cell transplant (HCT) and heralds an extremely poor prognosis. Cytokine induced memory-like (CIML) human NK cells have enhanced ability to recognize and kill leukemia targets. We initiated a phase I trial of CIML NK cells to treat patients with relapsed myeloid malignancies after haplo-HCT (NCT04024761). Transcriptional profiling analysis by scRNAseq were used to characterize the immunophenotypes of infused NK cells post cell adoptive transfer and as well as their interactions with tumor cells.

The Developmental Heterogeneity of Human Natural Killer Cells Defined by Single-cell Transcriptome

(Submitter supplied) The heterogeneity of human NK cells has not been fully-defined. Using single-cell RNA-sequencing technology, we find more human NK subsets than previously defined by cell surface markers. The transcriptome-based pseudotime analysis support that CD56bright NK cells are precursors of CD56dim NK cells with identification of a transitional stage. Our data significantly expand our understanding of the heterogeneity and development of human NK cells.
