Runx/CBFβ transcription factor complexes regulate Ccl5 expression through two novel enhancers to enhance tumor immunity [dataset 3]

(Submitter supplied) CCL5 (also known as RANTES) was originally known to be important to recruit memory T cells to infected sites, but it received much attention as an inhibitor to HIV’s entry into T lymphocytes of AIDS patients. Recently, it has been reported to be necessary to sustain Trm (Tissue-resident memory) T cells at local tissues as well as to play roles in manipulating tumor microenvironment by both host immunity and cancer cells. more...

Transcriptional regulation of adaptive NK and CD8 T cell responses [RNA-Seq]

(Submitter supplied) Natural killer (NK) cells are innate lymphocytes that possess features of adaptive immunity such as clonal expansion and generation of long-lived memory. Here we look at transcriptional profiles of NK cells throughout several time points during MCMV infection, ex-vivo cytokine stimulation, and/or deficiency of key transcription factors such as STAT4, STAT1, and Runx1. In addition, we profile parallel time points of MCMV-specific CD8 T cells during infection and memory formation.

Transcriptional and epigenetic regulation of adaptive NK cell responses [ChIP-Seq]

(Submitter supplied) Natural killer (NK) cells are innate lymphocytes that possess features of adaptive immunity such as clonal expansion and generation of long-lived memory. Interleukin (IL)-12 signaling through its downstream transcription factor STAT4 is required for the generation of memory NK cells following expansion, while type I interferon through STAT1 is important for NK cell expansion. Here, we identify gene loci that are highly enriched for STAT4 binding using ChIP-seq for STAT4 and the permissive histone mark H3K4me3 in activated NK cells. more...

Genome-wide maps of Cbfb variants bound regions in E12.5 lineage negative fetal liver cells and adult thymocytes.

(Submitter supplied) Multi-potent hematopietic progenitors must acquire thymus-homing capacity to initiate T lymphocyte development. Despite its importance, the transcriptional program underlying this process remains elusive. Cbfβ forms transcription factor complexes with Runx proteins, and here we sho that Cbfβ2, encoded by an RNA splice variant of the Cbfb gene, is essential for differentiation of IL7R+PIRhi thymic-homing progenitors in the mouse fetal liver. more...

Gene expression profiling of IL7R+PIR+ and IL7R-PIR- subsets in LIN-c-Kit+ E12.5 mouse fetal liver cells

(Submitter supplied) In order to find out taget genes of Runx/Cbfb2 complexes in PIR+IL7R+ subset. In particular, genes that would explain why the size of thymus was small by lack of Cbfb2. Upon finding of decresed IL7R+PIR+ subset, which was reported as thymus-homing progenitor, in Cbfb2 deficient embruos embryos, we focused on these population.

RNA-seq analyses of two intestinal dendritic cell subsets, CD103+CD11b- and CD103-CD11b+, from wild type and CbfbF/F:CD11c-Cre mice.

(Submitter supplied) We report phenotypes in gut dendritic cell development by loss of Runx/Cbfb transcription factor complexes. To further examine function of residual CD103+CD11b- and CD103-CD11b+ DCs, we performed RNA-seq analyses and compared gene expression signatures between control and Cbfb-deficient cells.

Cooperation between Constitutive and Inducible Chemokines Enables T-cell Engraftment and Immune Attack in Solid Tumors

(Submitter supplied) We investigated the role of chemokines in regulating T-cell accumulation in solid tumors. CCL5 and CXCL9 overexpression was associated with CD8+ T-cell infiltration in common solid tumors. T-cell infiltration required tumor cell-derived CCL5 and was amplified by IFN-dependent, myeloid cell-secreted CXCL9. Accordingly, CCL5-CXCL9 co-expression revealed immunoreactive tumors with prolonged survival and response to checkpoint blockade. more...

Core Binding Factor β Is Required For Group-2 Innate Lymphoid Cell Activation

(Submitter supplied) Group-2 innate lymphoid cells (ILC2) are tissue-resident, long-lived innate effector cells implicated in allergy and asthma. Upon activation, mature ILC2 rapidly secret large amounts of type-2 cytokines and other effector molecules. The molecular pathways that drive ILC2 activation are not well understood. Here we report that the transcriptional controller Core-binding factor β (CBFβ) is required for ILC2 activation. more...
