Transcriptional profiling of human hepatic NK cell subpopulations

(Submitter supplied) To identify potential functional roles of liver-resident NK cells we performed RNA sequencing on FACS sorted NK cell subpopulations from liver perfusates (n=5) and healthy blood controls (n=5). Full-length transcripts were sequenced from low-input samples using a modified version of the SMART-Seq2 protocol. Hepatic CD56brightCD16- and CD56brightCD16+ samples upregulate genes associated with tissue residency.

Gene expression profile at single cell level of mononuclear cells from the liver sinusoid

(Submitter supplied) This study identified a distinct liver sinusoidal NK-like CD56hiCD161-CD8+ T-cell population that expands in the liver with hepatitis B virus-associated chronic liver disease. CD56hiCD161-CD8+ T cells exert NKG2C-mediated NK-like effector functions even without TCR stimulation.

Human bone marrow resident natural killer cells have a unique transcriptional profile and resemble resident memory CD8+ T cells

(Submitter supplied) Human lymphoid tissues harbor, in addition to CD56bright and CD56dim natural killer (NK) cells, a third NK cell population: CD69+CXCR6+ lymphoid tissue (lt)NK cells. The function and development of ltNK cells remain poorly understood. In this study we performed RNA sequencing on the CD56bright and CD56dim NK cells (from bone marrow and blood), and the ltNK cells (from bone marrow). In addition, the blood derived CD56dim, and bone marrow derived ltNK cells were further subdivided into a NKG2A+ and NKG2A- fraction. more...

CD56dim NK cell gene signature in melanoma patients with better and worse clinical outcomes

(Submitter supplied) Natural killer (NK) cells are innate cytotoxic and immunoregulatory lymphocytes that have a central role in anti-tumor immunity and play a critical role in mediating cellular immunity in advanced cancer immunotherapies, such as dendritic cell (DC) vaccines. Our group recently tested a novel recombinant adenovirus-transduced autologous DC-based vaccine that simultaneously induces T cell responses against three melanoma-associated antigens for advanced melanoma patients. more...

Gene expression profile of CD56dimCD16+, CD56brightCD16- and activated CD56+CD16+ human NK subsets

(Submitter supplied) Pooled purified peripheral blood derived CD56dimCD16+ NK, CD56brightCD16- NK and in vitro activated CD56+CD16+ NK subsets obtained from 9 healthy donors were analyzed for gene expression pattern. Each pooled NK subset sample was hybridized in replicates (A and B). Keywords: other

Gene expression profile at single cell level of bulk liver allograft tissue isolated 12 months post-transplant in patients receiving regulatory dendritic cell therapy (STUDY) and reference standard-of-care patients (SOC)

(Submitter supplied) Regulatory dendritic cell therapy has shown potential in modulating the effector immune response after liver transplant in precliical models . We used single cell RNA sequencing (scRNA-seq) to analyze the differences in immune cell landscape of liver allograft at 12 months post-transplant in patients infused with regulatory dendritic cell therapy compared to standard-of-care control patients.

Single-cell RNA sequencing reveals the heterogeneity of liver-resident immune cells in human

(Submitter supplied) The liver plays a critical role in both immune defense and tolerance in the body. The liver-resident immune cells (LrICs) determine the immune properties, but the unique composition and heterogeneity of these cells are incompletely understood. Here, we dissect the diversity of LrICs by a comprehensive transcriptomic profiling using the unbiased single-cell RNA-sequencing (scRNA-seq). A total of 70, 706 of CD45+ immune cells from the paired liver perfusion, spleen and peripheral blood as references were profiled. more...
