Next Generation Sequencing Facilitates Quantitative Analysis of antidepressants stressed E. coli K-12 MG1655 Transcriptomes

(Submitter supplied) The goals of this study are to use Next-generation sequencing (NGS) to detect bacterial mRNA profiles of wild-type E. coli K-12 MG1655 and its mutants, and their mRNA response under the exposure of five antidepressants, including sertraline, duloxetine, bupropion, escitalopram and agomelatine. The concentrations were 50 mg/L for the treatment. For sertraline, in addition to 50 mg/L, 1 mg/L was also applied. more...

Next Generation Sequencing Facilitates Quantitative Analysis of E. coli K-12 LE392, P. putida KT2440, and Acinetobacter baylyi ADP1 Transcriptomes under various antidepressant exposure

(Submitter supplied) The goals of this study are to use Next-generation sequencing (NGS)to detect bacterial mRNA profiles of E. coli K-12 LE392, P. putida KT2440 and Acinetobacter baylyi ADP1 in response to various antidepressant concentrations for 2 h, in triplicate, using Illumina HiSeq 2500.The NGS QC toolkit (version 2.3.3) was used to treat the raw sequence reads to trim the 3’-end residual adaptors and primers, and the ambiguous characters in the reads were removed. more...

Dual effect: High NADH levels contribute to efflux-mediated antibiotic resistance but drive ROS lethality

(Submitter supplied) In light of the antibiotic crisis, emerging strategies to sensitize bacteria to available antibiotics should be explored. Several studies on the mechanisms of killing suggest that bactericidal antibiotic activity is enforced through the generation of reactive oxygen species (ROS lethality hypothesis). Here, we artificially manipulated the redox homeostasis of the model opportunistic pathogen Pseudomonas aeruginosa using specific enzymes that catalyze either the formation or oxidation of NADH. more...

Transcriptome profiles of Streptomyces coelicolor A3(2) wild type treated with kanamycin and iron

(Submitter supplied) We determine genes that responsible for iron induced resistance to kanamycin in Streptomyces coelicolor. Iron acts as a inducing agent for resistance to bactericidal antibiotics with concentration dependent manner. Identification of iron-dependent differentially expressed genes in wild-type by RNA-seq identified more than 100 genes. This series encompasses the RNA-seq data of our study.
