Real-time quantitative PCR analysis of Tomato stress response

(Submitter supplied) To extract RNA from shoots and roots, 3 weeks old control and treated plants were harvested at 2pm in the greenhouse. Total RNA was extracted using the AccuPrep® Universal RNA extraction kit (Bioneer, Daejeon, Korea) and treated with RNase-free DNase to remove DNA fragments (Qiagen, Hilden, Germany). One microgram of total RNA was used to synthesize cDNA with AccuPower® RT PreMix (Bioneer, Daejeon, Korea). more...

Real-time quantitative PCR analysis of tomato bZIP genes under biotic stress and plant hormone treatments

(Submitter supplied) We used Bio-Rad laboratories, lnc. PCR assay panel to analyze expression levels of bZIP genes in various biotci stress and plant hormone treatments. Tomato (Solanum lycopersicum) cv. Suhong 2003 were reared in growth chambers at 24 ± 1 °C with a photoperiod of 14 h light/10 h dark and light intensity of 200 μE m2 s-1. For analysis of gene expression, 4-week-old tomato plants were treated by foliar spraying with 100 μM methyl jasmonate (MeJA), 100 μM 1-amino cyclopropane-1-carboxylic acid (ACC), 100 μM SA or 0.1% ethanol as a control and samples were collected at indicated time points after treatment. more...

Real-time quantitative PCR analysis of tomato bZIP genes in various tissues

(Submitter supplied) We used Bio-Rad laboratories, lnc. PCR assay panel to analyze expression levels of bZIP genes in various tissues or organs. Tomato (Solanum lycopersicum) cv. Suhong 2003 were reared in growth chambers at 24 ± 1 °C with a photoperiod of 14 h light/10 h dark and light intensity of 200 μE m2 s-1. The roots, stem, leaves, flower, and fruits were collected from flowering plants for tissue expression analysis.

Transcriptome analysis of tomato seedlings infected by Pepino Mosaic Virus

(Submitter supplied) Pepino Mosaic Virus (PepMV) consists a major pathogenic threat in the greenhouses worldwide and has a devasting impact on tomato global production. PepMV belongs to the Potexvirus genus of the Flexiviridae family. PepMV has a viral RNA genome of approximately 6400 nucleotides long that contains five open reading frames (ORFs) and 4 major genotypes have been characterized. TomCr3, an indigenous virulent PepMV strain derived from the Chilean (CH2) genotype, was isolated in Crete, Greece and was used for the mechanical infection of Belladonna F1 hybrid tomato seedlings. more...

Transcriptome changes triggered by Hyaloperonospora parasitica arabidopsis Noco2 in WT and wrky72 mutants at 96 hpt

(Submitter supplied) Functional characterization of AtWRKY72 using Arabidopsis T-DNA insertion lines showed that this gene is important for basal defense to root-knot nematode (RKN) and Hyaloperonospora parasitica arabidopsis (Hpa), but not several tested R gene-mediated defenses. To profile transcriptional reprogramming associated with AtWRKY72-dependent basal defense we used Affymetrix ATH1 GeneChips representing ~24,000 Arabidopsis genes. more...

Transcription profiling of Arabidopsis plants overexpressing SlHsfA3

(Submitter supplied) In order to investigate the possible molecular mechanisms of SlHsfA3 function in plant growth and stress responses, we employed the RNA-seq approach to identify the genes with altered expression levels in the transgenic Arabidopsis overexpressing SlHsfA3. Processing of RNA samples on the Illumina HiSeq 2000 system yielded more than 24 million reads, each 100bp in length, encompassing 2.4Gb of sequence data for each sample which was then mapped to the reference genome. more...
