Serine/threonine kinase Tao-1 dominant negative mutation effect on pole cells

Analysis of pole cells overexpressing a dominant negative Tao-1 mutation. Tao-1 is a member of a conserved family of ste20-related serine/threonine kinases that regulate fundamental cellular processes including apoptosis. Results identify genes regulated by Tao-1 in pole cells undergoing apoptosis.

Overexpression of wild-type and dominant-negative form of tao-1

(Submitter supplied) Tao-1 is a member of evolutionarily conserved family of ste20-related serine/threonine kinases that regulate fundamental cellular processes including apoptosis. Drosophila tao-1 is expressed in pole cells during embryogenesis. To identify genes that act downstream of tao-1 in the pathway within pole cells, we compared the gene expression profiles of pole cells expressing dominant-negative form Tao-1 (D168A) and wild-type Tao-1 (D168D) by microarray analysis. more...

SMAUG Is a Major Regulator of Maternal mRNA Destabilization in Drosophila

(Submitter supplied) In animals, egg activation triggers a cascade of posttranscriptional events that act on maternally synthesized RNAs. We show that, in Drosophila, the PAN GU (PNG) kinase sits near the top of this cascade, triggering translation of SMAUG (SMG), a multifunctional posttranscriptional regulator conserved from yeast to humans. Although PNG is required for cytoplasmic polyadenylation of smg mRNA, it regulates translation via mechanisms that are independent of its effects on the poly(A) tail. more...

Expression data from third instar larval eye discs

(Submitter supplied) Expression of dE2F1 induces proliferation and apoptosis. We sought to perform an unbiased analysis of the effect of co-expression of miR-11 We used microarrays to identify differentially regulated genes following expression of dE2F1 and/or dme-miR-11 using the GMR-Gal4 driver

A male infertility mutation switches NANOS1 activity from anti-apoptotic to pro-apoptotic

(Submitter supplied) While Nanos-mediated maintenance of germ cells in Drosophila and mice has been related to regulation of apoptosis, the relevance of these findings to human physiology is uncertain. We show here that the NM_199461.4(NANOS1_v001):c.[100C>A;240_242del]; NM_199461.4(NANOS1_i001):p.[(Pro34Thr);(Ser83del)] double mutation, which has previously been associated with a lack of germ cells in the testes of infertile patients, causes NANOS1 to functionally switch from being anti-apoptotic to pro-apoptotic in the human male germ cell line TCam-2.
