Rheumatoid arthritis: synovial macrophages

Analysis of synovial fluid macrophages from 5 patients with rheumatoid arthritis (RA). The chronic inflammation and progressive joint destruction observed in RA are mediated in part by macrophages. Results provide insight into the molecular mechanisms underlying RA pathogenesis.

Interferon beta-1a-induced changes in gene expression in PBMCs derived from CIS patients

(Submitter supplied) IFNβ, an effective therapy against relapsing-remitting (RR) multiple sclerosis (MS) is naturally secreted during the innate immune response against viral pathogens. The objective of this study was to characterize the immunomodulatory mechanisms of IFNβ targeting innate immune response and their effects on DC-mediated regulation of T-cell differentiation. We found that IFNβ−1a in-vitro treatment of human monocyte-derived dendritic cells (DCs) induced the expression of TLR7 and the members of its downstream signaling pathway, including myeloid differentiation factor 88 (MyD88), IL-1R-associated kinase (IRAK)4, and TNF receptor-associated factor (TRAF)6, while it inhibited the expression of IL-1R. more...

MEK1/2 inhibitors unlock the constrained interferon response in macrophages through IRF1 signaling

(Submitter supplied) Analysis of bone-marrow derived macrophages (BMDMs) stimulated with single or combinatorial TLR agonist and MEK1/2 inhibitor in wild-type and Irf1-/- macrophages. Results provide insights into the versatile but yet uncharacterized roles of IRF1 in TLR-MEK1/2-ERK MAPK signaling.

Epidermal Growth Factor Receptor inhibition triggers Type 1 Interferon signature in human keratinocytes

(Submitter supplied) The Epidermal Growth Factor Receptor (EGFR)/ligand system is centrally involved in multiple homeostatic functions of the epithelia. Epithelial cells are the primary targets of humanized antibodies and small molecule inhibitors against this system, whereby the constellation of skin-specific side effects of these drugs stems from a profound disturbance of keratinocyte biology. So far, the molecular mechanisms underlying these toxic events have been investigated only broadly. more...
