Non-phosphorylated FTY720 effect on immortalized microglia HMO6 cell line

Analysis of immortalized brain HMO6 microglia treated with FTY720-non-P, a non-phosphorylated form of FTY720 (a synthetic analog of sphingosine). Results provide insight into the molecular mechanisms responsible for triggering FTY720-non-P-induced apoptosis of HMO6.

Non-phosphorylated FTY720 induces apoptosis of human microglia by activating SREBP2

(Submitter supplied) A synthetic analog of sphingosine named FTY720 (Fingolimod), phosphorylated by sphingosine kinase-2, interacts with sphingosine-1-phosphate (S1P) receptors expressed on various cells. FTY720 suppresses the disease activity of multiple sclerosis (MS) chiefly by inhibiting S1P-dependent egress of autoreactive T lymphocytes from secondary lymphoid organs, and possibly by exerting anti-inflammmatory and neuroprotective effects directly on brain cells. more...

RNA-seq for understanding astrocyte S1P1 signaling

(Submitter supplied) Astrocytic S1P1 has been previously shown to be essential for FTY720 (an orally delivered drug for MS) efficacy (CNS), in addition to its well-known immunological mechanisms of action. FTY720-P (an analogue of S1P) is a functional antagonist to S1P1. TetTag c-Fos reporter mice displayed temporal and spatial GFP-labeling of astrocytic nuclei under experimental autoimmune encephalomyelitis (EAE) pathology. more...

RNA-seq for understanding effects of vitamin B12 removal on astrocyte culture

(Submitter supplied) Astrocytes express a vitamin B12 uptake receptor, CD320/TCblR, that is regulated by S1P1 signaling. In search of genes controlled under B12 deficiency in astrocytes, we employed RNA-seq using RNA of astrocytes cultured in normal or B12 deficient media.

Hippocampal expression data from FTY720- and vehicle-treated SCID mice following fear consolidation testing

(Submitter supplied) FTY720/Fingolimod, an FDA-approved drug for treatment of multiple sclerosis, has beneficial effects in the CNS that are not yet well understood, independent of its effects on immune cell trafficking. Here we show that FTY720 enters the nucleus where it is phosphorylated by sphingosine kinase 2 (SphK2) and nuclear FTY720-P that accumulates there, binds and inhibits class I histone deacetylases (HDACs) enhancing specific histone acetylations. more...

Expression analysis of Kasumi-1 cells treated with or without FTY720

(Submitter supplied) The clinical outcomes of M2 subtype Acute Myeloid Leukemia (AML-M2) with t(8;21) are poor. Here we report that FTY720 (Fingolimod), a sphingosine analogue and an FDA approved drug for treatment of multiple sclerosis, showed great antitumorigenic activity against Kasumi-1 cell line, xenograft mouse model and leukemic blasts isolated from AML-M2 with t(8;21) patients. Primary investigation indicated that FTY720 caused cell apoptosis through caspase and protein phosphatase 2A (PP2A) activation. more...

Inflammatory stress signaling via NF-kB alters accessible cholesterol to upregulate SREBP2 transcriptional activity in endothelial cells

(Submitter supplied) There is a growing appreciation that a tight relationship exists between cholesterol homeostasis and immunity in leukocytes, however, this relationship has not been deeply explored in the vascular endothelium. Endothelial cells (ECs) rapidly respond to extrinsic signals, such as tissue damage or microbial infection, by upregulating factors to activate and recruit circulating leukocytes to the site of injury and aberrant activation of ECs leads to inflammatory based diseases, such as multiple sclerosis and atherosclerosis. more...
