Interleukin-21 effect on STAT3-deficient naïve B cells from autosomal dominant hyper IgE syndrome patients

Analysis of naïve B cells purified from peripheral blood of AD-HIES patients with STAT3 loss-of-function mutation and then cultured with CD40 ligand alone or together with interleukin-21. Results provide insight into IL-21R/STAT3 signaling induced genes underlying naïve B cell differentiation.

Identification of IL-21-induced STAT3 dependent genes in human B cells

(Submitter supplied) IL-21 induces B cell activation, and differentiation into antibody-secreting plasmablasts in vitro. This process is abolished by loss-of function mutations in STAT3 We used microarrays to identify genes that are induced by IL-21 in a STAT3-dependent manner

Gene expression in TFH cells

(Submitter supplied) After activation, CD4+ helper T (Th) cells differentiate into distinct effector subsets. Although chemokine (C-X-C motif) receptor 5-expressing T follicular helper (Tfh) cells are important in humoral immunity, their developmental regulation is unclear. Here we show that Tfh cells had a distinct gene expression profile and developed in vivo independently of the Th1 or Th2 cell lineages. Tfh cell generation was regulated by ICOS ligand (ICOSL) expressed on B cells and was dependent on interleukin-21 (IL-21), IL-6, and signal transducer and activator of transcription 3. more...

Cytokine receptor modulation by interleukin-2 broadly regulates T helper cell lineage differentiation

(Submitter supplied) T helper (Th) cells control host defense to pathogens. IL 12R expression is required for Th1, IL-4Rα for Th2, and IL-6Rα/gp130 for Th17 differentiation to allow responsiveness to IL-12, IL-4, and IL-6, respectively. IL-2 via STAT5 controls Th2 differentiation by regulating the Th2 cytokine gene cluster and Il4ra expression. Here we show that IL-2 regulates Th1 differentiation, inducing STAT5-dependent IL-12Rβ2 and T-bet expression, with impaired human Th1 differentiation when IL-2 was blocked. more...

Transcriptome Analysis of Follicular Helper T cells, Central Memory Precursors, and Terminal Effector Th1 Cells from Wild Type and Stat3fl/fl Cd4cre mice

(Submitter supplied) Purpose: To compare the transcriptomes of activated CD4 T effector cell populations in the presence and absence of STAT3 at 8 days post-infection using high-throughput RNA sequencing analysis. Methods: Cell sorting of the populations was done using the markers Ly6c and PSGL-1

PD biomarkers for IL-22Fc

(Submitter supplied) The goal is to identify an IL-22Fc specific gene signature in human intestinal epithelial cells in order to support PD biomarkers for IL-22Fc. We have identified IL-22Fc specific activity in HT-29 cells as secreted acute phase proteins only in the presence of IL-1β. Therefore, HT-29 cells (from gCell) will be cultured with IL-22, IL-1β, IL-6 (as a pSTAT3 activation control) as well as IL-22 + IL-1β and IL-6 + IL-1β. more...

Bulk RNA-seq and ATAC-seq of mouse uterine tissue-resident natural killer cells.

(Submitter supplied) Uterine natural killer(NK) cells play a crucial role in pregnancy by promoting spiral artery remodeling and secreting fetal growth factors.Uterine trNK cells before pregnancy and during pregnancy (gd6.5, gd8.5 and gd11.5) were sorted, then applied into bulk RNA-sequence to reveal the features at different timepoints. Similarily, uterine trNK cells with different treatment in vitro were also sorted for bulk RNA-sequence to identify the role of IL-21-STAT3 signaling. more...
