Pathogen-associated molecular-pattern curdlan effect on interleukin-2 deficient GM-CSF myeloid dendritic cells

Analysis of GM-CSF myeloid dendritic cells (GM-DCs) isolated from interleukin-2 (IL-2) knockouts and activated with curdlan, a pathogen-associated molecular pattern (PAMP). GM-DCs secrete cytokines such as IL-2, upon PAMP activation. Results provide insight into role of IL-2 signaling in DCs.

Dendritic cell-derived IL-2 promotes apoptosis of terminally mature cells via a novel autocrine signaling pathway

(Submitter supplied) Dendritic cells (DCs) are crucial for sensing pathogens and triggering immune response. GM-CSF myeloid dendritic cells (GM-DCs) secrete several cytokines including IL-2 upon activation by pathogen associated molecular pattern (PAMP) ligands. DC IL-2 has been shown to be important for innate and adaptive immune responses, however its importance in DC physiology has never been demonstrated. This is due to ambiguity in expression of the CD122 subunit of the IL-2 trimeric receptor complex crucial for signaling. more...

Synergism between curdlan and GM-CSF in mouse dendritic cells

(Submitter supplied) A simultaneous engagement of different pathogen recognition receptors provides a tailor made adaptive immunity for an efficient defence against distinct pathogens. For example, cross talk of TLR and c-type lectin signalling effectively shapes distinct gene expression patterns by integrating the signals at the level of NF-κB. Here, we extend this principle to a strong synergism between the Dectin-1 agonist, curdlan, and an inflammatory growth factor, GM-CSF. more...

Cytokine-dependent dendritic cell differentiation in the splenic microenvironment

(Submitter supplied) The dendritic cell (DC)-derived chemokine CCL17, a ligand of CCR4, has been shown to promote various inflammatory diseases such as atopic dermatitis, atherosclerosis, and inflammatory bowel disease. Under steady state conditions, and even after systemic stimulation with lipopolysaccharide, CCL17 is not expressed in resident splenic DC as opposed to CD8-CD11b+ lymph node (LN) DC, which produce large amounts of CCL17, in particular after maturation. more...
