Effect of sulforaphane on gene expression in Spodoptera exigua and Trichoplusia ni

(Submitter supplied) Cruciferous plants produce sulforaphane (SFN), an inhibitor of nuclear histone deacetylases (HDACs). In humans and other mammals, the consumption of SFN alters enzyme activities, DNA-histone binding, and gene expression within minutes. However, the ability of SFN to act as a HDAC inhibitor in nature, disrupting the epigenetic machinery of insects feeding on these plants, has not been explored. Here, we investigate the effect of SFN, as well as the pharmaceutical HDAC inhibitor Trichostatin A (TSA), consumed in the diet in the generalist grazer Spodoptera exigua and in the co-evolved specialist feeder Trichoplusia ni. more...

Expression patterns of TnAV-6a1 genes in the combination of fat body, Epidermis and tracheal matrix tissues compared to hemolymph viral vesicles.

(Submitter supplied) Purpose: To compare the TnAV-6a1 variant infected Trichoplusia ni larval transcriptome of hemolymph viral vesicles versus those of other major larval tissues specifically, the fat body, epidermis and tracheal matrix (referred to as FET tissues).

Formation of the CenH3-deficient holocentromere in Lepidoptera avoids actie chromatin

(Submitter supplied) Despite the essentiality for faithful chromosome segregation, centromere architectures are diverse among eukaryotes and embody two main configurations: mono- and holocentromeres, referring respectively to localized or unrestricted distribution of centromeric activity. Of the two, some holocentromeres offer the curious condition of having arisen independently in multiple insects, most of which have lost the otherwise essential centromere-specifying factor CenH3 (first described as CENP-A in humans). more...

Eukaryote-Specific Insertion Elements Control Human ARGONAUTE Slicer Activity

(Submitter supplied) We solved the crystal structure of human ARGONAUTE1 (hAGO1) bound to endogenous 5'-phosphorylated guide RNAs. To identify structural changes that evolutionarily rendered hAGO1 inactive, we compared our structure with published structures of guide-RNA-containing and cleavage-active hAGO2. Aside from mutation of a catalytic tetrad residue, proline residues at positions 670 and 675 in hAGO1 introduce a kink in the cS7 loop and thereby forming a convex surface within the hAGO1 nucleic-acid-binding channel near the inactive catalytic site. more...
