Treatment protocol |
After incubation for 24 hours at 37 ºC in a humidified incubator with 5% CO2, epi4 cells were stimulated with 1 μM capsaicin for 4 hours.

Growth protocol |
Epi4 cells were seeded into a 12-well culture plate at a concentration of 2 × 105 cells in Epilife medium (Cascade Biologics, Portland, OR) supplemented with S7 (Cascade Biologics).

Extraction protocol |
Total RNA was isolated from the cells using TRI Reagent (Molecular Research Center, Cincinnati, OH, USA) following the manufacturer's recommendations.

The RNA samples were amplified and labeled with Cy3 by use of the Quick Amp Labeling Kit (Agilent Technologies, Santa Clara, CA, USA), followed by RNAeasy column purification (QIAGEN, Valencia, CA). Dye incorporation and cRNA yield were checked with the NanoDrop ND-1000 Spectrophotometer (Agilent Technologies).

Hybridization protocol |
The Cy3-labelled cRNA was fragmented at 60°C for 30 minutes using Agilent fragmentation buffer (Agilent). On completion of the fragmentation reaction, the samples were competitively hybridized onto the Agilent 4 × 44K whole human genome oligo microarray slide (Agilent) for 17 hours at 65C°in a rotating Agilent hybridization oven.

Scan protocol |
Slides were scanned immediately after washing on the Agilent DNA Microarray Scanner using one color scan setting.
