Hybridization protocol |
Description : Slides were UV-crosslinked with 6000μJ and prehybridized for one hour in 3xSSC, 0.1% SDS and 1% blocking agent (Roche). Slides were boiled for two minutes prior to drying. Labeled cDNA was hybridized to slides under lifter slips (Erie Glass) overnight at 65?C in a hybridization buffer containing 3xSSC, 0.1% SDS, 20mM Hepes, 13.6 micrograms tRNA and 40 micrograms polyA RNA. All dye handling and hybridizations were conducted in an ozone free room.
