Treatment protocol |
Adult (8-10-week- old), pathogen-free male mice (C57BL6/J from Jackson Laboratories, Bar Harbor, Maine) were housed one to a cage for a week for acclimatization before experimental dosing. Mice that contained disrupted α-synuclein (α-synuclein knockout mice, stock number 003692) were treated similarly. Age-matched wild type C57BL6/J mice that contained α-synuclein served as the control animals for knockout experiments. The animals were housed under standard conditions of temperature and humidity with a 12 h light/dark cycle and fed a standard pellet diet and water ad libitum. Animals were treated with normal saline (control), paraquat (10mg/kg, ip) and Maneb (30mg/kg, ip) twice a week for 6 weeks.

Total RNA was isolated from the nigrostriatal tissue of three independent triplicates using the TRIzol (Ambion, Life Technologies, Grand Island, NY) and RNeasy mini kit columns with in-column DNAse treatment (Qiagen, Valencia, CA).

Starting with 500ng of total RNA, Cy3 labeled cRNA was produced according to manufacturer’s protocol.

Hybridization protocol |
For each sample, 3 ug of Cy3 labeled cDNA was hybridized for 17 hours in a rotating hybridization oven.

Scan protocol |
Slides were washed and then scanned with an Agilent Scanner.
