Growth protocol |
Hepatoblasts were maintained on a type IV collagen-coated culture plate in a medium consisting of a 1:1 mixture of DMEM/Ham’s F-12 with L-glutamine, HEPES and sodium pyruvate (Wako) supplemented with 10% fetal bovine serum (vol/vol) (Nichirei Bioscience), 1 µg/ml insulin (Wako), 100 nM dexamethasone (Sigma-Aldrich), 10 mM nicotinamide (Sigma-Aldrich), 50 µM β-mercaptoethanol (Nacalai Tesque), 50 ng/ml human recombinant hepatocyte growth factor (HGF) (Peprotech) and 20 ng/ml epidermal growth factor (EGF) (Peprotech).

Extraction protocol |
Total RNA was extracted using the RNeasy Mini Kit (Qiagen) and was assessed by Agilent 2100 Bioanalyzer and the RNA 6000 LabChip Kit (Agilent Technologies).

The array data were analyzed using GeneSpring software. Gene expression values were normalized by excluding low-signal intensity data and percentile shifts.
