Extraction protocol Sorted cells from 5 animals were pooled. The total RNA was isolated from cells by using RNeasy Plus Mini Kit (Qiagen) with additional purification and concentration with RNA Clean& Concentrator (Zymo Research).

Label protocol 100 ng of total RNA was mixed with Reporter CodeSet and Capture ProbeSet from nCounter Mouse Inflammation V2 panel according to the instructions at nCounter® XT Assay User Manual (NanoString tecnology) User Manual
 
Hybridization protocol Samples were hybridized for 18 h at 65C according to the instructions at nCounter® XT Assay User Manual (NanoString tecnology)
Scan protocol After hybridization samples were placed in the prep station of nCounter FLEX analysis system, there hybridized samples were purified and immobilized in a sample cartridge (cartridge ID 207911400919) according to standard protocol. Then the sample cartridge was scanned on the nCounter FLEX digital analyzer (Scanner ID 1410C0216)

Data processing The raw data were processed in nSolver Analysis Software 4.0. We used CodeSetContent(Housekeeping) standard normalization
