Effect of Tunicamycin on Bovine Capillary Endothelial Cells

(Submitter supplied) GPT inhibitor Tunicamycin develops ER stress causing anti-angiogenic response in breast tumor microvasculature due to unfolded protein response-mediated apoptosis. cDNA microarray identified 123 and 454 differentially regulated genes with 10 genes overlapping between 3h and 32 h of Tunicamycin treatment. Alg-2 expression is inconsistent but not the Dpms. Evidences support that Tunicamycin completely destroys DPMS activity in capillary endothelial cells without affecting its protein or the mRNA levels but by knocking down the phosphorylation. more...

Effects on the hepatic transcriptome due to caloric restriction are not altered by milking frequency

(Submitter supplied) The ruminant liver has multiple roles in the dairy cow and many of these are crucial in nutrient supply during lactation. Reduced feed intake alters the expression of many genes and pathways in the liver, inducing a period of negative energy balance. Once-daily milking is a management strategy to reduce the effects of periods of negative energy balance so the objective of this study was to determine if once-daily milking altered hepatic gene transcription during a period of negative energy balance induce by caloric restriction. more...

Inferring the in vivo cellular program of developing bovine muscle from expression data

(Submitter supplied) We outline an in vivo cellular program of bovine longissimus muscle development inferred from expression data from 60 days post conception to 3 months postnatal. Analytic challenges included changes in cellular composition, ambiguous ‘diagnostic’ markers of cell type and contrasts between cattle human and mouse myogenesis. Nevertheless, the expression profiles of the myosin isoforms support slow and fast muscle fibres emanating from primary and secondary myogenesis respectively, while expression of the prenatal myosin subunits is down regulated prior to birth. more...

Transcriptional profiling of cumulus cells differentiation throughout oocyte competence acquisition in the bovine ovarian follicles.

(Submitter supplied) Somatic cells surrounding the oocyte were sampled at the following stages: developmentally incompetent or poorly competent prophase I oocytes (NC1 oocytes), developmentally competent prophase I oocytes (C1 oocytes), and developmentally competent metaphase II oocytes (C2 oocytes). NC1 cumulus cells (CC) were sampled from immature calf oocytes, C1 samples from immature cow oocytes, and C2 samples from in vivo matured cow oocytes.

Colony stimulating factor 2 causes changes in the transcriptome of the bovine preimplantation embryo

(Submitter supplied) The objective of this study was to identify changes on the CSF2 embryo transcriptome that make them more able to become a blastocyst and more likely to survive after transfer. Embryos were treated with CSF2 at day 5 after fertilization and at day 6 morulae and blastocysts were select for the microarray analysis. A total of 160 genes that had a 1.5-fold difference in expression and a significance of P ≤ 0.05 could be annotated. more...

Transcriptional profiling of bovine endometrial tissue at day 17 of the reproductive cycle

(Submitter supplied) Over the past three decades, there has been a significant decline in dairy cattle fertility. A large proportion of pregnancy losses are believed to occur during the pre-implantation period, when the developing embryo is elongating rapidly and signaling its presence to the maternal system. The molecular mechanisms that prevent progression of the estrous cycle and allow the allogenic embryo to survive within the maternal environment are not well understood. more...
