TCDD effect on regenerating fin: time course

Analysis of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) exposed caudal fins at various time points up to 5 days post amputation. TCDD prevents caudal fin regeneration. Results provide insight into the pathways required for fin regeneration and the mechanisms of TCDD toxicity.

1, 3, or 5 Day Post Amputation Vehicle or TCDD Exposed

(Submitter supplied) Adult zebrafish can completely regenerate their caudal fin following amputation. This complex process is initiated by the formation of an epithelial would cap over the amputation site by 12 hours post amputation (hpa). Once the cap is formed, mesenchymal cells proliferate and migrate from sites distal to the wound plane and accumulate under the epithelial cap forming the blastemal structure within 48 hpa. more...

2,3,7,8-tetrachlorodibenzo-p-dioxin effect on cardiovascular development

Analysis of gestational day (GD) 17.5 fetal hearts isolated from pregnant C57BL/6Ns treated at GD 14.5 with up to 6.0 ug/kg 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). TCDD, a cardiovascular teratogen, inhibits cardiomyocyte proliferation, a process that peaks between GD 14.5 to 17.5.

Comparative genomic analysis between adult and larval fin regeneration

(Submitter supplied) Zebrafish have the remarkable ability to regenerate body parts including the heart, spinal cord and fins by a process referred to as epimorphic regeneration. Recent studies have illustrated that similar to adult zebrafish, early life stage-larvae also possess the ability to regenerate the caudal fin. A comparative genomic analysis was used to determine the degree of conservation in gene expression among the regenerating adult caudal fin, adult heart and larval fin. more...

Gene Expression Profiles of Intact and Regenerating Zebrafish Retina

(Submitter supplied) Purpose: Investigate the molecular determinants of retinal regeneration in adult vertebrates by analyzing the gene expression profiles of control and post-lesion retina of adult zebrafish, a system that regenerates following injury. Methods: Gene expression profiles of zebrafish retina and brain were determined with DNA microarray, RT-PCR, and real-time quantitative PCR analyses. Damaged retinas and their corresponding controls were analyzed 2-5 days post-lesion (acute injury condition) or 14 d post-lesion (cell regeneration condition). more...

1Day Post Amputation Vehicle or Beclomethasone Exposed

(Submitter supplied) Zebrafish caudal fin regeneration is an established model to study tissue regeneration. In order to identify novel molecular signaling pathways critical for regeneration, we developed a rapid throughput in vivo regeneration assay. We screened a 2000 member structurally diverse small molecule library, followed by assessment of regenerative progression at three days post amputation. A cluster of glucocorticoids was identified among the “positive hits”. more...

(Submitter supplied) Confluent C3H10T1/2 cells stimulated to differentiate using IDMB (1uM BRL49653, 1uM Dexamethasone, 0.5uM IBMX, 10ug/mL Insulin). Expression was examined 24 hours after IDMB treatment. Additionally, the effects of addition of TCDD 48 hours prior to IDMB treatment, and EGF addition concurrent with IDMB treatment were examined. Hormone-induced adipogenesis suppressed by 2,3,7,8-tetrachlorodibenzo-p-dioxin and EGF

Analysis of differentiated C3H10T1/2 fibroblasts treated with 10 nM 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) and/or 10 nM EGF. Prior to treatment, cells induced to differentiate into adipocytes with IDMB hormone. EGF increases effectiveness of TCDD in suppressing hormone-induced adipogenesis.

(Submitter supplied) This study investigates the effects of the aryl hydrocarbon receptor (AhR) ligands TCDD, PCB126 and PeCDF; the non-AhR ligand PCB153 and the binary mixture PCB126/PCB153 on hepatic gene expression in female sprague dawley rats. Rats were treated with toxicological equivalent doses of TCDD (100ng/kg), PeCDF (200ng/kg), PCB126 (1000ng/kg) and PCB153 (1000ug/kg) 5 days a week for 13 weeks. Single-cell resolution of MET- and EMT-like programs in osteoblasts during zebrafish fin regeneration

(Submitter supplied) We performed single-cell RNA-sequencing (scRNA-seq) to elucidate osteoblastic transcriptional programs during zebrafish caudal fin regeneration. We show that osteoprogenitors are enriched with components associated with epithelial-to-mesenchymal transition (EMT) and its reverse, mesenchymal-to-epithelial transition (MET). Trajectory analyses indicate osteoblastic cells solely expressed EMT components, or transiently expressed components for MET before EMT. more...
