Host cell response to HIV-1 Vpr-induced cell cycle arrest: time course

Analysis of cell lines expressing wild-type or mutant HIV-1 Vpr protein at various timepoints following doxycycline induction. Vpr advances HIV pathogenesis by inhibiting the cell cycle at G2/M. Results identify changes in cellular gene expression associated with Vpr-mediated cell cycle arrest.

Effects of inducible HIV-1 Vpr protein on cellular gene expression

(Submitter supplied) Three 293 cell-derivative cell line were constructed using commercially available Invitrogen Flp-In TREx 293 cells. The cell lines express wild type HIV-1 Vpr and two mutant Vprs, F72A/R73A and R80A. The mutant Vprs are defective in the Vpr cell cycle arrest phenotype. The cell lines were characterized for their ability to express Vpr at the RNA and protein levels and functionally for the ability of the inducibly expressed Vprs to arrest cell cycle progression at G2/M. more...

The expression of microRNA Induced by HIV-1 Infection

(Submitter supplied) To investigate whether and what miRNAs expression might be regulated by HIV-1 challenge, we analyzed the miRNA expression profile of MT4 cells inoculated with HIV-1-GFP for 48h using an array-based miRNA profiling and the infectivity was over 90% by analyzing with flow cytometer.

Single cell analysis on the HIV-infected kidney cells

(Submitter supplied) HIV-vpr expression was mainly detected in tubule cells after infection. By comparing the HIV-infected and wild type kidney cells on single cell level, a strong shift was observed in the PT S3 segment cell population. Transcription factor analysis revealed Trp53 playing important regulation role in HIV infected cells.
