A549 lung pneumocytes cell line infected with wildtype and deletion mutation PAK strain Pseudomonas aeruginosa

(Submitter supplied) The bacterial type III secretion system (TTSS) is dedicated to directly effect host cell pathways by pathogens. In order to identify the key host responses of the various parts of the TTSS, we utilized expression profiling of a lung pneumocytes cell line, A549, exposed to various isogenic mutant strains of Pseudomonas aeruginosa PAK. We have devised a novel filtering method to isolate the key responses to the effector proteins as well as the translocation machinery. more...

Lung pneumocyte response to Pseudomonas aeruginosa type III secretion system mutants

Analysis of A549 lung pneumocytes after infection with Pseudomonas aeruginosa (PA) PAK mutants deleted for various components of the type III secretion system (TTSS). Mutants in TTSS effectors and translocators studied. PA causes serious respiratory infections in cystic fibrosis patients.

Global expression profiling of airway epithelial cells infected with Pseudomonas aeruginosa and the rsmA mutant

(Submitter supplied) Global expression profiling of airway epithelial cells infected with Pseudomonas aeruginosa and the rsmA mutant. Recent work in our laboratory investigated the impact of RsmA on a number of airway epithelial cell phenotypes including actin depolymerization, cytotoxicity and invasion. These cellular phenotypes were influenced by the positive effect of RsmA on the expression of the TTSS in P. aeruginosa (Mulcahy et al. more...

Airway epithelial cell response to Pseudomonas aeruginosa rsmA mutant infection

Analysis of airway epithelial cells infected with Pseudomonas aeruginosa PAO1 or the rsmA mutant. RsmA is a small RNA-binding protein that plays a role in the regulation of virulence-related genes in P. aeruginosa. Results provide insight into the influence of RsmA-regulated functions on the host.

Expression profiles of hrpRS- and hrpL- mutants

(Submitter supplied) Pseudomonas syringae pv. tomato DC3000 is a model pathogen infecting tomato and Arabidopsis plants. Genes encoding the type III secretion system and substrate proteins (collectively called TTSS genes) of this bacterium are induced in plants and in minimal medium (MM). The induction of TTSS genes is mediated by HrpL, an alternative sigma factor recognizing the hrp box in the promoter of TTSS genes. The transcription of hrpL is activated by HrpR and HrpS, two homologous DNA binding proteins encoded by the hrpRS operon. more...
