caArray_cho-00156: Gene Expression in Ovarian Cancer Reflects Both Morphology and Biological Behavior

(Submitter supplied) Biologically and clinically meaningful tumor classification schemes have long been sought. Some malignant epithelial neoplasms, such as those in the thyroid and endometrium, exhibit more than one pattern of differentiation, each associated with distinctive clinical features and treatments. In other tissues, all carcinomas, regardless of morphological type, are treated as though they represent a single disease. more...

ANALYSIS OF GENE EXPRESSION IN EARLY-STAGE OVARIAN CANCER (2)

(Submitter supplied) Gene expression was analysed in 68 stage I ovarian cancers and 15 borderline samples using microarrays. Tumors were obtained directly at surgery and immediately frozen in liquid nitrogen until analysis. Glass arrays containing 16000 genes were used. Data was interpreted by unsupervised and supervised analysis. All tumors had a wild-type p53. Unsupervised analysis identified eight major clusters one of which was statistically associated to overall survival, grading and histotype (cluster C) and another with grading and histotype (cluster D). more...

Immune-activation as a therapeutic direction for patients with high-risk ovarian cancer based on gene expression signature (2)

(Submitter supplied) Advanced-stage ovarian cancer is one of the most lethal gynecologic malignancies. To improve prognosis of patients with ovarian cancers, a predictive biomarkers leading to personalized treatments are required. In this large-scale cross-platform study of six microarray datasets consisting of 1054 ovarian cancer patients, we developed a novel risk classification system based on a 126-gene expression signature for predicting overall survival by applying elastic net7 and 10-fold cross validation to a Japanese dataset A (n = 260). more...

Gene expression data from non-malignant fallopian tube epithelium and high grade serous carcinoma.

(Submitter supplied) The purpose of this study was to identify molecular alterations potentially involved in predisposition to adnexal serous carcinoma (SerCa) in the non-malignant fallopian tube epithelium (FTE) of BRCA1/2-mutation carriers, given recent evidence implicating the distal FTE as a common source for SerCa. Conventional renal cell carcinomas

(Submitter supplied) Our goal was to identify gene expression features, using comprehensive gene expression profiling, that correlate with survival in conventional renal cell carcinomas (cRCCs). We profiled 177 cRCCs using high-density cDNA microarrays. Unsupervised hierarchical clustering analysis segregated cRCC into five gene expression subgroups. Expression subgroup was correlated with survival in long-term follow-up and was independent of grade, stage, and performance status. more...

Gene Expression Profiles of a Progression Model of Human Ovarian Cancer

(Submitter supplied) We have earlier reported the establishment of a comprehensive in vitro panel of 19 isogenic cell lines from a patient with Grade IV serous adenocarcinoma (Bapat et al. Cancer Research. 2005). In this study, one of the cell lines termed as A4 at early passes (less than ~20) was assessed as being non-tumorigenic since it failed to exhibit anchorage independence growth in soft agar and form tumors in nude mice. more...

A molecular classification of papillary renal cell carcinoma

(Submitter supplied) BACKGROUND: Despite the moderate incidence of papillary renal cell carcinoma (PRCC), there is a disproportionately limited understanding of its underlying genetic programs. There is no effective therapy for metastatic PRCC, and patients are often excluded from kidney cancer trials. A morphological classification of PRCC into Type 1 and Type 2 tumors has been recently proposed, but its biological relevance remains uncertain. more...

Papillary renal cell carcinoma classification

Analysis of renal tumors from 34 papillary renal cell carcinoma (PRCC) cases. Results identify 2 molecular signatures that correlate with the morphologic classification of PRCC. Molecular class type 1 tumors correlate with excellent survival, while type 2 tumors correlate with poor survival.

Two distinct gene signatures identify malignant Neuroblast and Schwannian stromal cells of Neuroblastic Tumors

(Submitter supplied) Tumor tissue heterogeneity is a well known feature of several solid tumors. Neuroblastic Tumors (NTs) is a group of paediatric cancers with a great tissue heterogeneity. Most of NTs are composed of undifferentiated, poorly differentiated or differentiating neuroblastic (Nb) cells with very few or absent Schwannian stromal (SS) cells: these tumors are grouped as Neuroblastoma (Schwannian stroma-poor). more...

DNA profiling of serous ovarian and Fallopian tube carcinomas with array CGH and MLPA

(Submitter supplied) Primary serous ovarian carcinoma (OVCA) and serous Fallopian tube carcinoma (FTC), both belonging to the BRCA-linked tumour spectrum, share many properties and are treated similarly. However, a detailed molecular comparison has been lacking. We hypothesized that comparative genomic studies of serous OVCAs and FTCs should point to gene regions critically involved in their tumorigenesis. Array comparative genomic hybridization (array CGH) analysis indicated that serous OVCAs and serous FTCs displayed common but also more distinctive patterns of recurrent changes. more...

Gene Experssion Profiling-Based Identification of Molecular Subtypes in Stage IV Melanoma with Different Clinical Outcome (validation set)

(Submitter supplied) Purpose: The incidence of malignant melanoma is increasing worldwide in fair-skinned populations. Melanomas respond poorly to systemic therapy, and metastatic melanomas inevitably become fatal. Although spontaneous regression, likely due to immune defense activation, rarely occurs, we lack a biological rationale and predictive markers in selecting patients for immune therapy. Experimental Design: We performed unsupervised hierarchical clustering of global gene expression data from stage IV melanomas in 57 patients. more...
