Gene expression profiles of Hodgkin’s lymphoma cell lines with different sensitivity to cytotoxic drugs

(Submitter supplied) By using high-density DNA microarrays, we analyzed the gene-expression profile of Hodgkin's lymphoma cell lines. Furthermore, we tested the sensitivity of these cell lines for cytotoxic drugs (cisplatin, etoposide, melphalan) and compared the gene-expression profile of chemotherapy-resistant and -sensitive cell lines. Classical Hodgkin lymphoma shows epigenetic features of an abortive plasma cellular differentiation: expression of PCM vs. B-cell lines

(Submitter supplied) Background Epigenetic changes are involved in the extinction of the B-cell gene expression program of classical Hodgkin lymphoma. However, little is known regarding epigenetic similarities between classical Hodgkin lymphoma and plasma cell myeloma cells, both of which share an extinction of the gene expression program of mature B-cells. Design and methods Global histone H3 acetylation patterns were determined in cell lines derived from classical Hodgkin lymphoma, plasma cell myeloma and B-cell lymphoma by chromatin immunoprecipitation and subsequent hybridization onto promoter tiling arrays. more...

Epigenetic reprogramming in relapsed childhood ALL

(Submitter supplied) Reversing gene expression signatures in relapsed patient may restore chemosensitivity. We demonstrate that the histone deacetylase inhibitor vorinostat not only reprograms the aberrant gene expression profile of relapsed blasts but is synergistic when applied prior to chemotherapy in primary patient samples and leukemia cell lines

Increased Protein Processing Gene Signature in HDACi-resistant cells predicts response to proteasome inhibitors.

(Submitter supplied) Epigenetic modifying enzymes are commonly mutated in diffuse large B cell lymphoma (DLBCL). Importantly, genetics abnormalities lead to inactivation of HAT, which tilt the balance in favor of decreased protein acetylation in DLBCL cells. This suggests that protein acetylation regulation is an important factor in DLBCL pathogenesis and a potential target for therapy. We developed resistant cell lines to the histone deacetylase inhibitor (HDACi) vorinostat, in order to better define molecular mechanisms of action of HDACi in lymphoma cells. more...

Transcriptome analysis of KM-H2 cells with different Rhodamin 123 eflux capacity

(Submitter supplied) Rhodamin 123 is a dye which can be used to detect the activity of ABC transporters. We observed that after staining of KM-H2 Hodgkin lymphoma cells with Rhodamin 123, part of the cells rapidly eliminated the dye, while another part of the cells retained the dye for a longer time. We compared the transcriptome of KM-H2 Hodgkin lymphoma cells with high Rhodamin 123 efflux capacity and KM-H2 cells with low Rhodamin 123 efflux capacity.
