Regulation of gene expression by RORgamma in Th17 cells

(Submitter supplied) We observed significant growth inhibition of melanoma tumor in RORγ-/- mice, suggesting an important role for the T helper 17 cell (TH17) pathway in tumor immunity. Though RORgamma t (RORγt) plays a critical role in the development of IL-17-secreting Th17 cells, the role of other genes remained an open question. To explore the expression of unknown genes with anti-tumor properties, gene expression analysis was performed.

transferred T helper cell sorted from mice

(Submitter supplied) Mice were injected s.c. in the front of the abdomen with 1 × 106 B16-OVA tumor cells. At 10 days after tumor injection, mice (5/group) were treated with adoptive transfer of 2.5 × 106 Th1, Th9 or Th17 cells, followed by i.v. injection of 2.5 × 105 peptide-pulsed BMDC. Total RNA was extracted with RNeasy Mini kit (Qiagen) from CD45.1+CD4+ Th cells sorted from spleens of tumor-bearing mice 12 days after transfer.

Pulmonary interstitial macrophages are required to mediate the pro-tumorigenic effects of IL-9

(Submitter supplied) We demonstrate that deficiency of IL-9 signaling inhibits lung tumor growth in multiple models. In defining the IL-9-responsive cells in the lung tumor models, we found IL-9 regulates lung heterogeneity by inhibiting alveolar macrophage and promoting the expansion of both CD11c+ and CD11c- interstitial macrophage populations.
