Expression data from canine B-cell lymphomas

(Submitter supplied) Pet dogs may provide a useful large-animal model of spontaneously occuring non-Hodgkin lymphoma, but complete molecular characterization is lacking. We used gene expression microarrays to molecularly characterize canine lymphoma, and found similarities between canine B-cell lymphoma and human diffuse large B-cell lymphoma.

Translocations Activating IRF4 Identify a Subtype of Germinal-Center-Derived B-cell Lymphoma Affecting Predominantly Children and Young Adults

(Submitter supplied) Background: Germinal center B-cell (GCB) lymphomas are common in children and adults. The prognosis strongly depends on age. Subgroups of GCB-lymphomas are characterized by chromosomal translocations affecting immunoglobulin (IG) loci leading to oncogene deregulation. Methods: Novel IG translocation partners were cloned within the network “Molecular Mechanisms in Malignant Lymphomas” (MMML) by long-distance inverse polymerase chain reaction. more...

Comparative Transcriptional Analysis of Canine and Human Diffuse Large B cell Lymphoma (DLBCL). Molecular Signatures of NF-κB Pathway Activation and Sub-Classification of Canine DLBCL.

(Submitter supplied) In this study, we present the first comparison of global transcriptional changes taking place in canine lymphoma and human diffuse large B-cell lymphoma (DLBCL), with particular reference to the nuclear factor-kappaB (NF-κB) pathway. Microarray data generated from lymph nodes diagnosed with canine DLBCL and healthy lymph nodes were used for differential expression analysis, co-expression analysis and pathway analysis, and compared with analysis of publicly available microarray data from human healthy and DLBCL lymph nodes. more...

The impact of transgenic Uchl1 on gene expression in germinal center B-cells from ImHABCL6 mice

(Submitter supplied) This study seeks to understand the mechanisms behind enhanced lymphomagenesis observed in ImHABCL6/Uchl1 mice compared with ImHABCL6 alone. As the lymphomas arise from germinal center (GC) B-cells, we reasoned that transgenic Uchl1 altered the gene expression patterns in GC B-cells from these animals. We therefore isolated pre-malignant GC B-cells and examined the gene expression patterns to identify pathways affected by the addition of Uchl1.

MicroRNA profiling of primary cutaneous large B-cell lymphomas

(Submitter supplied) High-throughput sequencing of primary cutaneous follicle center lymphoma (PCFCL), primary cutaneous diffuse large B-cell lymphoma, leg type (PCLBCL-LT) and in vitro activated peripheral blood B-cells. We performed high-throughput sequencing analysis on frozen tumor biopsies from 19 cases of PCFCL and PCLBCL-LT to establish microRNA profiles. Cluster analysis of the complete microRNome could not distinguish between the two subtypes, but 16 single microRNAs were found to be differentially expressed.

Smc3 regulates B-cell transit through germinal centers and restricts their malignant transformation (single-cell RNA-seq)

(Submitter supplied) During the germinal center (GC) reaction, B-cells undergo extensive redistribution of cohesin complex and 3D re-organization of their genomes. Yet, the significance of cohesin and architectural programming in the humoral immune response is unknown. Herein we report that conditional homozygous deletion of cohesin subunit Smc3 abrogated GC formation, yet in marked contrast, Smc3 haploinsufficiency induced GC hyperplasia, skewing of GC polarity and impaired plasma cell differentiation. more...

CD137 (4-1BB) ligand mediates CD4 T-cell immuno-surveillance of pre- lymphoma B cells in a lymphoma-prone mouse model

(Submitter supplied) Aberrant expression of the proto-oncogene BCL6 is a driver of tumorigenesis in diffuse large B cell lymphoma (DLBCL). Mice overexpressing BCL6 from the B cell-specific immunoglobulin heavy chain μ intron promoter (Iμ-Bcl6 Tg/+ ) develop B cell lymphomas with features typical of human DLBCL. B cell lymphoma development in these mice is tightly controlled by T cells; however, the mechanisms of this immune surveillance are poorly understood. more...

Gene expression profiling and mutation analysis of B cell lymphomas arising in mice expressing NIK and BCL6 transgenes

(Submitter supplied) In this study, by RNA sequencing we generated the gene expression profile (GEP) of each mouse lymphoma. By comparing the GEP of a lymphoma to that of normal B cells at physiological differentiation stages, we classified the lymphomas to either ABC- or GCB-type of DLBCL. Analysis of RNA sequencing data also led to identification of various mutations in these lymphomas.

Dimethyl fumarate induces ferroptosis and impairs NF-κB/STAT3 signaling in DLBCL

(Submitter supplied) Multi-agent chemotherapy still represents the first-line standard-of-care treatment for diffuse large B-cell lymphoma (DLBCL), the most common form of lymphoma in adults. However, the clinicopathological and molecular heterogeneity of DLBCLs poses a major challenge in their successful therapy. At least two major subtypes, i.e. germinal center B-cell-like (GCB) and the aggressive activated B-cell-like (ABC) DLBCL, which differ both in their gene expression profile and in their mutation patterns, have been identified. more...

Gene Expression Profiling reveals a close relationship between Follicular lymphoma Grade 3A and 3B, but distinct profiles of Follicular Lymphoma Grade 1 and 2

(Submitter supplied) Since follicular lymphoma (FL) grade 3A often coexist with a FL1/2 component a linear progression model of FL1, FL2 and FL3A has been developed. FL3B, on the other hand, is supposed to be more closely related to diffuse large B-cell lymphoma (DLBCL) and both FL3B and DLBCL are often simultaneously present in one tumor (DLBCL/FL3B). To obtain more detailed insight into FL progression, comprehensive analysis of a well-defined set of FL1/2 (n=22), FL3A (n=16), FL3B (n=6), DLBCL/FL3B (n=9) and DLBCL (n=45) was performed using gene expression profiling, immunohistochemical staining and genetic analysis by fluorescence in situ hybridization (FISH).
