Transcriptome analysis of Wnt3a-treated triple-negative breast cancer cell lines.

(Submitter supplied) We analyzed the transcriptome of two different triple negative breast cancer (TNBC) cell lines to define a comprehensive list of Wnt target genes. Cells were treated with Wnt3a for 6h, 12h or 24h. We found up-regulated and down-regulated genes in response to Wnt3a treatment. They are involved in the Wnt pathway itself, and also in TGFß, p53 and Hedgehog pathways. Thorough characterization of these novel potential Wnt target genes may reveal new regulators of the canonical Wnt pathway. more...

FZD7 Plays a Critical Role in Triple Negative Breast Cancer Proliferation

(Submitter supplied) Breast cancer is genetically and clinically heterogeneous. Triple negative cancer (TNBC) is a subtype of breast cancer usually associated with poor outcome and lack of benefit from target therapy. A pathway analysis in a microarray study was performed using TNBC compared with non-triple negative breast cancer (non-TNBC). Overexpression of several Wnt pathway genes, such as frizzled homolog 7 (FZD7), Low density lipoprotein receptor-related protein 6 (LRP6) and transcription factor 7 (TCF7) has been observed in TNBC. more...

Triple negative breast cancer

Analysis of triple negative breast cancer (TNBC) tumors. TNBC is usually associated with poor outcome, and is characterized by lack of estrogen and progesterone hormone receptors and lack of overexpression of HER2. Results provide insight into the molecular basis of tumorigenesis of TNBC.

The WNT target SP5 negatively regulates WNT transcriptional programs in human pluripotent stem cells

(Submitter supplied) The WNT/β-catenin signaling pathway is a prominent player in many developmental processes, including gastrulation, anterior-posterior axis specification, organ and tissue development and homeostasis. Here, we use human pluripotent stem cells (hPSCs) to study the dynamics of the transcriptional response to exogenous activation of the WNT pathway. We describe a mechanism involving the WNT target gene SP5 that leads to termination of the transcriptional program initiated by WNT signaling. more...

Gene expression in Rspo2 and/or Wnt3a treated C57MG mouse mammary epithelial cells

(Submitter supplied) R-spondins (Rspos) comprise a family of four secreted proteins that have important roles in cell proliferation, cell fate determination and organogenesis. Rspos typically exert their effects by potentiating the Wnt/β-catenin signaling pathway. To systematically investigate the impact of Rspo/Wnt on gene expression, we performed a microarray analysis using C57MG mouse mammary epithelial cells treated with recombinant Rspo2 and/or Wnt3a.

Gene expression profiling induced by high dose ionizing radiation on MDA-MB-231 cells

(Submitter supplied) Radiation therapy (RT) remains a vital component of the curative multimodality therapy for many types of cancer including breast cancer (BC). Even if great number of BC patients receive RT, not all of them report benefits, due to radioresistance activation depending also by hormone receptor status. We analyze and compare molecular responses, in term of gene expression profile (GEP) changes, induced by high dose of Ionizing Radiation (IR), in MDA-MB-231 with ER-/PR-HER2- receptor status. more...

(Submitter supplied) Triple negative breast cancer is an aggressive phenotypic breast cancer characterized by ER negative, PR negative and Her2 negative immunohistochemistry status. We embarked on a study to explore the transcriptome of Kenyan TNBC patients and identify potential biomarkers specific to Kenyan population. The transcriptome sequencing of tumors from Kenyan TNBC patients and comparisons with African American and Caucasian TNBC transcriptomes revealed several interesting targets and dysregulated pathways.
