(Submitter supplied) We report the RNA sequencing of both non-restimulated and restimulated (using anti CD3/CD28 cross-linking) antigen-experienced (CD44+) mouse lung CD8 T cells 21 days post X31 influenza A infection subsetted by integrin expression of CD49a and CD103. Expression analysis of human naïve and memory CD8 T cells at resting stage and following short-term stimulation

(Submitter supplied) Memory T cells (TM) play a prominent role in protection and auto-immunity due to their ability to mount a more effective response than naïve T cells (TN). However, the molecular mechanisms underlying enhanced functionality of TM are not well defined, particularly in human TM. We examined the global gene expression profiles of human CD8+ TN and TM before and after stimulation. To compare the expression profile of CD8 T cells from mice vaccinated with Ad85A via the intranasal (IN) or intradermal (ID) routes isolated from lungs or spleen.

(Submitter supplied) In Balb/c mice, the presence of effector CD8 T cells in lungs after intra-nasal (IN) immunization with replication deficient recombinant Adenovirus expressing the 85A antigen from Mycobacterium tuberculosis (MTb) (Ad85A) correlates with protection against aerosol MTb infection. In order to identify differentially expressed transcripts which contribute to protection by IN immunized lung CD8 T cells, total RNA from CD8 T cells isolated from target tissue (lungs) and lymphoid organ (spleen ) after a protective (IN) and non-protective (intradermal, ID) immunization regime will be analyzed in a whole-genome microarray study.

Genome-wide profiling of early and late CD8 memory T cells expressing CD62L

(Submitter supplied) Memory CD8+ P14 cells were generarted through adoptive transfer and infection with LCMV armstrong. Then, early memory (after 30 - 45 days) and late memory (after 8 months) cells were sort purified based on CD62L expression. Genome-wide expression profiles were captured for early and late memory cells with high and low levels on CD62L using Affymetrics arrays to show their molecular and functional differences.

Characterization of CD8 T cells during acute LCMV infection using scRNA-seq

(Submitter supplied) During a viral infection, CD8 T cells encounter a myriad of antigenic and inflammatory signals of variable strength, which sets off each individual T cell on a unique differentiation trajectory. However, the developmental path for each of these cells will ultimately lead to one of only two potential outcomes after clearance of the infection—death or survival and development into memory CD8 T cells. How this cell fate decision is made remains incompletely understood. In this study, we explore the dynamic transcriptional changes during effector and memory CD8 T cell differentiation at the single cell level.
