Transcriptome analysis of rhesus monkey germinal vesicle and failed-to-mature oocytes

(Submitter supplied) Oocyte maturation defects are one contributing factor in unexplained female infertility. Failure of some oocytes to undergo germinal vesicle breakdown or progress to second meiotic metaphase in response to an ovulatory stimulus can limit the number of high quality oocytes available for assisted reproduction. Understanding the molecular mechanisms that underlie oocyte maturation failure may lead to novel methods for increasing the number of high quality oocytes available for assisted reproduction, as well as providing new insight into the basic biology of oocyte maturation. more...

Transcriptome analysis of immature and matured human oocytes from patients of young and advanced maternal age

(Submitter supplied) Study Question: What effects do maternal age and oocyte maturation stage have on the human oocyte transcriptome that may be associated with oocyte developmental potential? Summary Answer: Although polyadenylated transcript abundance changes during human oocyte maturation irrespective of age, young (YNG) and advanced maternal age (AMA) metaphase II (MII) oocytes exhibit divergent transcriptomes. What is known already: Maternal age and maturation stage affect oocyte polyadenylated transcript abundance in human oocytes. more...

The impact of ageing on the transition of the euploid human GV oocytes to in vivo matured MII oocytes

(Submitter supplied) Background: Early embryonic development is governed by maternal transcripts stored within the oocyte during oogenesis. Transcriptional activity of the oocyte ultimately dictates its developmental potential and may be influenced by maternal age, resulting in reduced competence of oocytes derived from women of advanced age, compared with the young. In the current study, RNA-Seq was used to perform transcriptome profiling of human GV and MII oocytes derived from young and advanced maternal age women. more...

The transcriptome of human oocytes is related to age and ovarian reserve.

(Submitter supplied) Although it is well established that the ovarian reserve diminishes with increasing age, and that a woman’s age is correlated to lower oocyte quality, the interplay of a diminished reserve and age on oocyte developmental competence is not clear. After maturation, oocytes are mostly transcriptionally quiescent, and developmental competence prior to embryonic genome activation (EGA) relies on maternal RNA and proteins. more...

Good quality day 7 embryos display disturbed cellular functions during post-hatching development when oocytes were matured under lipotoxic conditions: novel insights from a bovine embryo-transfer model

(Submitter supplied) Objective: To study post-hatching development after transfer of morphologically good quality embryos that survived in vitro maturation under lipolytic conditions using a well-established and recognized bovine model. Design: prospective study with a crossover design. Setting: research laboratory. Animals: 8 healthy multiparous non-lactating Holstein cows used in repeated embryo transfers. Intervention(s): Bovine cumulus oocyte complexes (COCs) were in vitro matured in the presence of either 1) a combination of elevated pathophysiological concentration of palmitic acid (PA) and physiological concentrations of stearic acid (SA) and oleic acid (OA) (HIGH PA), or 2) physiological concentrations of PA, SA and OA (BASAL). more...

Chromosome constitution in embryos generated after in vitro maturation in a culture system supplemented with cAMP modulators

(Submitter supplied) Ten volunteers underwent 13 research IVM cycles. Immature oocytes were retrieved after mild ovarian stimulation with gonadotropins but no hCG ovulation trigger, in vitro matured and fertilized using ICSI. Oocyte maturation rate in vitro was 47% (94/199), the mean fertilization rate was 71% (67/94) and 37% (25/67) of mature oocytes developed into a morphologically good quality embryo on day 3 after ICSI, after which the blastomeres were analysed by single-cell aCGH. more...

Small RNA profiling of human cumulus cells and oocytes

(Submitter supplied) Cumulus cells are biologically distinct from other follicular cells and perform specialized roles, transmitting signals within the ovary and supporting oocyte maturation during follicular development. The bi-directional communication between the oocyte and the surrounding cumulus cells is crucial for the acquisition of oocyte competence. Using Illumina/deep-sequencing technology, we dissected the small RNAome of pooled human mature MII oocytes and cumulus cells.

Specific processing of meiosis related transcript is linked to final maturation in human oocytes

(Submitter supplied) Human meiosis in oocytes entails a fine regulation of the transcriptome to support late oocyte growth and early embryo development, both key to reproductive success. Currently, little is known about the co- and post-transcriptional mRNA processing mechanisms regulating the last meiotic passages, which contribute to transcriptome complexity and influence translation rates. We analyzed gene expression changes, splicing and pre-mRNA processing in an RNA sequencing set of 40 oocytes at different meiotic maturation stages, both in vivo ovulated and matured in vitro. more...

Storage time does not modify the gene expression profile of cryopreserved human metaphase II oocytes

(Submitter supplied) Study question: Does storage time impact on transcriptome of slowly frozen cryopreserved human metaphase II (MII) oocytes? Summary answer: For the first time, we demonstrate that the length of cryostorage has no effect on the gene expression profile of human metaphase II oocytes. What is known already: Oocyte cryopreservation is a largely-used technique in IVF for storage of surplus oocytes, as well as for fertility preservation (i.e., women undergoing gonadotoxic therapies) and oocyte donation programs. more...

Presence of aggregates of smooth endoplasmic reticulum in MII oocytes affects oocyte development competence: a molecular-based evidence

(Submitter supplied) The oocytes presenting SERa are considered dysmorphic and have been associated with reduced embryological outcome and an increased risk of congenital anomalies. Therefore, in 2011 the Alpha/ESHRE Consensus strongly recommended not to inseminate MII oocytes displaying SERa (SERa+). Not all Centers follow this recommendation and some Authors reported that SERa+ can lead to healthy newborns. Moreover, no experimental data about the effect of SERa presence on the oocyte molecular status are available. more...

Granulosa cell expression data from five different time points before and up to 36 hours after induction of ovulation in women receiving fertility treatment

(Submitter supplied) The process of ovulation includes oocyte meiotic maturation, follicle rupture and transformation of the follicle into a corpus luteum. These events are initiated by the midcycle surge of gonadotropins and require the coordinated regulation of thousands of genes. The aim of the study was to monitor the changes in granulosa cell gene expression across five different time points during the first 36 hours of ovulation until follicle rupture, in order to increase our understanding of the events of human ovulation. more...

Porcine oocytes matured in a chemically defined medium are transcriptionally active

(Submitter supplied) The statement that fully-grown porcine oocytes are transcriptionally quiescent is not as strong as before. Currently, we know that there is a difference in the transcription profile of germinal vesicle (GV) and metaphase II (MII) oocytes. The goal of our study was to compare the transcription profile of GV, germinal vesicle breakdown (GVBD), metaphase I (MI), and metaphase (MII) oocytes cultivated in a chemically defined, FLI medium. more...

Gene Expression Profiles in Immature and In vitro matured bovine Oocytes

(Submitter supplied) In cattle, almost all fully grown vesicle stage oocytes (GV) have the ability to resume meisos, develop to Metaphase II stage (MII), support fertilization and progress through the early embryonic cycles in vitro. Yet without intensive selection, the majority fail to develop to the blastocyst stage. Using the Affymetrix Bovine Genome Array, global mRNA expression analysis of immature (GV) and in vitro matured (IVM) bovine oocytes was carried out to characterize the transcriptome of the bovine oocyte and to identify the key pathways associated with oocyte meiotic maturation and developmental potential.
