A network of transcriptome modules demonstrates mechanistic pathways of both virus induced and non-viral asthma exacerbations in children [blood]

(Submitter supplied) We leverage a systems-scale network analysis approach to demonstrate repertoires of cellular transcriptional pathways underlying loss of asthma control, and show how these pathways differ in viral associated and non-viral exacerbations.

Virus-induced inflammatory gene networks underlying asthma exacerbations in vivo

(Submitter supplied) Global patterns of gene expression was profiled in nasal lavage samples obtained from asthmatic children during an acute Picornavirus-induced exacerbation and 7-14 days later. Gene coexpression network analysis and prior knowledge was employed to reconstruct the wiring diagram of the underlying gene networks.

Acute Picornavirus-induced exacerbation in asthmatic children: nasal lavage cells

Analysis of paired nasal lavage samples obtained from asthmatic children during an acute Picornavirus-induced asthma exacerbation in vivo and 7-14 days later. Severe episodes can be life-threatening. Results provide insight into molecular mechanisms underlying virus-induced asthma exacerbations.

Gene expression data from severe asthmatic children: PBMC profiles during acute exacerbation versus convalescence

(Submitter supplied) Severe asthma exacerbations in children requiring hospitalisation are typically associated with viral infection, and occur almost exclusively amongst atopics, but the significance of these comorbidities is unknown. We hypothesised that underlying interactions between immunoinflammatory pathways related to responses to aeroallergen and virus are involved, and that evidence of these interactions is detectable in circulating cells during exacerbations. more...

Gene expression profiling of nasal swabs samples from children with asthma or wheezing exacerbations.

(Submitter supplied) Nasal swab specimens were collected from children who presented to the emergency department with an acute exacerbation of asthma or wheeze. Samples were also collected from control subjects. Convalescent/quiescent samples were collected from children who were followed-up at least 6 weeks after an acute exacerbation of asthma or wheeze. Gene expression was profiled on microarrays.

Gene Expression Patterns in Peripheral Blood Mononuclear Cells Associated with Asthma Exacerbation Attack

(Submitter supplied) A large, prospective, non-interventional study was designed to study gene expression changes in peripheral blood mononuclear cells (PBMCs) associated with asthma exacerbations over the course of a year. PBMC samples were collected from subjects at the time of the study visits defined as 1) Quiet: during stable disease at 3 month intervals, 2) Exacerbation: during a 14 day period of deteriorating asthma and 3) Follow-up: within 14 days after cessation of an exacerbation. more...

LPS binding protein and activation signatures are upregulated during asthma exacerbations in children

(Submitter supplied) Asthma exacerbations in children are associated with respiratory viral infection and atopy, resulting in systemic immune activation and infiltration of immune cells into the airways. The gene networks driving the immune activation and subsequent migration of immune cells into the airways remains incompletely understood. Cellular and molecular profiling of PBMC was employed on paired samples obtained from atopic asthmatic children (n = 19) during acute virus-associated exacerbations and later during convalescence. more...

Phenotypic responses of differentiated asthmatic human airway epithelial cultures to rhinovirus

(Submitter supplied) We report the application of RNA sequencing technology for high-throughput profiling of gene expression responses to human rhinovirus infection at 24 hours in air-liquid interface human airway epithelial cell cultures derived from 6 asthmatic and 6 non-asthmatic donors. RNA-seq analysis identified sets of genes associated with asthma specific viral responses. These genes are related to inflammatory pathways, epithelial remodeling and cilium assembly and function, including those described previously (e.g. more...
