Characterization of early transcriptomic changes associated with hepatitis B virus exposure in human and macaque immune cell populations

(Submitter supplied) Chronic hepatitis B virus (HBV) infection affects close to 300 million individuals worldwide, representing one of the major etiological factors for the development of cirrhosis and hepatocellular carcinoma (HCC). At the molecular level, the mechanisms behind chronic HBV infection are based on the persistence of the viral genome as an episomal structure termed covalently closed circular DNA (cccDNA), and the evasion of both innate and adaptive immune responses. more...

Multi-parametric analysis of human livers reveals variation in intrahepatic inflammation across phases of chronic hepatitis B infection

(Submitter supplied) Chronic HBV is clinically categorized into 4 phases by a combination of serum HBV DNA levels, HBeAg status and alanine aminotransferase (ALT): immunotolerant (IT), immune-active (IA), inactive carrier (IC) and HBeAg-negative hepatitis (ENEG). Immune and virological measurements in the blood have proven useful but are insufficient to explain the interrelation between the immune system and the virus since immune dynamics differ in the blood and liver. more...

Transcriptomic profiling of peripheral blood NK cells of chronic HBV, HCV and HIV patients

(Submitter supplied) Background: NK cells during chronic viral infection have been well studied over the last decade. We performed an unbiased next-generation RNA-sequencing approach to identify commonalities or differences of the effect of HIV, HCV and HBV viremia on NK cell transcriptomes. Methods: Using cell sorting, we obtained CD3-CD56+ NK cells from blood of 6 HIV, 11 HCV, and 32 HBV infected and untreated patients. more...

Hepatitis B virus deregulates cell cycle to promote viral replication and a premalignant phenotype

(Submitter supplied) Hepatitis B virus (HBV) infection is a major health problem worldwide and chronically infected individuals are at high risk of developing cirrhosis and hepatocellular carcinoma (HCC). The molecular mechanisms whereby HBV causes HCC are largely unknown. By using a biologically relevant system of HBV infection of primary human hepatocytes (PHHs), we studied how HBV perturbs gene expressions and signaling pathways of infected hepatocytes, and whether these effects are relevant to productive HBV infection and HBV-associated HCC. more...

Characteristics of impaired dendritic cell function in patients with hepatitis B virus infection

(Submitter supplied) Dendritic cells are antigen-presenting cells with a central role in host immune response. This study analyzed gene expression and dendritic cell function in hepatitis B virus (HBV) patients and functions impaired due to HBV, and identified the genes related to these functions. Peripheral blood mononuclear cells from 23 HBV patients and 9 healthy controls were utilized. Mononuclear cells were isolated from side-scatter and forward-scatter gating using fluorescence-activated cell sorting (FACS), the lineage-negative HLA-DR-positive fraction was extracted, and was further divided into CD123-positive plasmacytoid DCs (pDCs) and CD11c-positive myeloid DCs (mDCs). more...
