Caffeine effect on TOR complex 1 mutant: dose response

Analysis of Tor1p mutant treated with up to 9 mM caffeine. Tor1p is a component of TOR complex 1 (TORC1), a regulator of growth in eukaryotic cells. I1954V and W2176R substitutions in Tor1p confer resistance to caffeine. Results provide insight into the function and activity of TORC1.

Time-course studies of three cell wall interfering drugs on gene expression in yeast

(Submitter supplied) Caffeine is a natural purine analog that elicits pleiotropic effects, which ultimately lead to cell death by a mechanism that is still largely uncharacterized. This drug can activate the PKC1-MAPK cell integrity pathway, as shown by phosphorylation of Mpk1 kinase. Transcript profiling of Candida albicans tor1 kinase mutants

(Submitter supplied) This study analyses the effects of truncation of the gene encoding Tor1 kinase in Candida albicans. The study involves a mutant tetO-TOR1-Del381 which has the N terminal regulator HEAT repeats deleted compared to a full length tetO-TOR1 strain and a heterozygous TOR1/tor1 control. Cell were grown in vitro in YEPD broth at 30˚C. In shut off experiments, doxycycline (30 µg.ml) was added to cultures to shut off the tetO promoter and inhibit Tor1 expression.
