Blastocyst dormancy and activation comparison

Comparison of gene expression in dormant and active blastocysts. Dormancy induced by ovariectomy and maintained by progesterone injections. Dormant blastocysts activated by estradiol-17 beta injections. Results provide insight into delayed implantation.

Global gene expression analysis identifies molecular pathways distinguishing blastocyst dormancy and activation

(Submitter supplied) Delayed implantation occurs in many mammals, but the underlying mechanisms that direct this process remain largely unknown. In mice, ovariectomy prior to the preimplantation ovarian estrogen secretion on day 4 of pregnancy initiates blastocyst dormancy, which can be maintained for more than 200 days by continued progesterone Keywords: other

Discovery of candidate genes and pathways in the endometrium regulating ovine blastocyst growth and conceptus elongation

(Submitter supplied) Establishment of pregnancy in ruminants requires blastocyst growth to form an elongated conceptus that produces interferon tau, the pregnancy recognition signal, and initiates implantation. Blastocyst growth and development requires secretions from the uterine endometrium. An early increase in circulating concentrations of progesterone (P4) stimulates blastocyst growth and elongation in ruminants. This study utilized sheep as a model to identify candidate genes and regulatory networks in the endometrium that govern pre-implantation blastocyst growth and development. more...

Analysis of the gene expression pattern of bovine blastocysts at three stages of development

(Submitter supplied) Blastocyst formation is a primordial event of pre-implantation development since it is required for pregnancy establishment and progression. The blastocyst plays a pivotal role because it is the stage at which the embryo is transferred and starts coordinated cross-talks with the mother. It is also the terminal step of pre-implantation developmentbefore transfer; it reflects all stresses the embryo may have faced during the process of in-vitro treatment. more...

Transcriptomic profile reveals the global remodeling of blastocyst for implantation

(Submitter supplied) Implantation of the blastocyst into the uterus is a developmental milestone in mammalian embryonic development, which is the gateway for further development of fetus. At this time, remodeling of the blastocyst to an implantation competent status, which is known as blastocyst activation, is indispensable for embryo implantation. This RNA sequence data is from the Dormant, Reactivating and Reactivated blastocyst.

Transcriptome changes at the initiation of elongation in the bovine conceptus

(Submitter supplied) Temporal changes in the embryo transcriptome between the blastocyst stage (Day 7) and initiation of elongation (Day 13) differ between in vivo- and in vitro-derived embryos and are reflective of subsequent developmental fate. The aim of this study was to examine the temporal changes in transcriptional profile as the embryo develops from a spherical blastocyst on Day 7 to an ovoid conceptus at the initiation of elongation on Day 13 and to highlight differences in these temporal gene expression dynamics between in vivo- and in vitro-derived blastocysts which may be associated with embryonic survival/mortality using the bovine Affymetrix microarray.

Gene expression profiles at the day 5 blastocyst implantation and interimplantation sites of the hamster [human array]

(Submitter supplied) Current knowledge of the molecular regulation of the blastocyst implantation event has been largely derived from studies in the mouse that requires ovarian estrogen for initiation of the implantation event. However, there are species such as the hamster, guinea pig, pig, horse, rhesus monkey and perhaps the human where the blastocyst implantation event initiates only in the progesterone-primed uterus. more...

m6A RNA methylation orchestrates transcriptional dormancy during paused pluripotency [SLAM-seq]

(Submitter supplied) Development is often assumed to be a sequential unfolding of genetic programs towards increased complexity and occurring with a very stereotypical timing. However, embryos across all lineages of metazoans can enter reversible states of developmental pausing in response to adverse environmental conditions. In mammals, pausing manifests as a delayed implantation of the blastocyst, the source of embryonic stem cells (ESCs). more...
