Gene expression of Grx4 regulated strains in the Ustilago maydis FB2 background

(Submitter supplied) Gene expression of FB2 wt in minimal medium with 2 percent arabinose or glucose after 24h of growth was compared with the conditional promoter regulated strain Pcrg1:grx4 (#55). Glucose is a repressor of this promoter while arabinose acts as inducer. Grx4 was initially pre-depleted during growth on MM with glucose over night. Afterwards cells were transferred to MM with either glucose or arabinose and cells were collected after 24h. more...

Global gene expression of post-senescent telomerase negative ter1Δ strain of Ustilago maydis

(Submitter supplied) The dimorphic fungus Ustilago maydis transits from unicellular sporidia to multicellular mycelium and accomplish its life cycle in planta with teliospore formation. Mutations in either of telomerase core subunits, trt1 or ter1 cause deficiencies or inability to produce galls and teliospores when crossed to a wild-type strain. Here, the global transcriptome analysis of two ter1Δ survivor strains of U. more...

The mRNA stability factor Khd4 defines a specific RNA regulon for membrane trafficking in the pathogen Ustilago maydis [RNA-Seq]

(Submitter supplied) Fungal pathogens depend on sophisticated gene expression programs for successful infection. A crucial component is RNA regulation mediated by RNA-binding proteins. In the biotrophic fungal plant pathogen Ustilago maydis, loss of the multi KH domain containing RBP, Khd4, causes aberrant cell morphology, disturbed hyphal growth and impaired virulence. To investigate the role of Khd4 in the polar growth of infectious hyphae, we established the RNA-editing based hyperTRIBE method to detect in vivo mRNA targets. This technique is especially suited to investigate proteins of high molecular weight or low expression that are difficult to purify (McMahon et al., 2016, Xu et al., 2018, Rahman et al., 2018). more...

Systematic characterization of Ustilago maydis sirtuins shows Sir2 as a modulator of pathogenic gene expression

(Submitter supplied) To investigate the role of Sir2 controlling gene expression during U. maydis pathogenesis, sir2 was deleted and overexpressed with Ppit2 promoter. RNA-Seq analysis were performed during axenic growth or filament formation in PD-Charcoal plates comparing Δsir2 mutan and wild type strains. Additionally RNA-Seq analysis were performed at 3 days post infection comparing Ppit2:sir2 >1c overexpression mutant and wild type strains.

A combination of organic acids and glucose prime a late stage of fungal biotrophy

(Submitter supplied) We show that combinations of organic acids and glucose trigger phenotypes associated with the late stage of biotrophy for the maize pathogen Ustilago maydis. These phenotypes include the expression of a set of effectors normally observed only during biotrophic development as well as the formation of melanin associated with sporulation in plant tumors.

Time series transcriptional profiling of Ustilago maydis SG200 infecting Zea mays.

(Submitter supplied) Goals: Comparing the infection between Ustilago maydis SG200 with the wild-type strain FB1xFB2 previously published Methods: Comparative RNASeq analysis between U. maydis SG200 and U. maydis FB1xFB2 at three timepoints (axenic, 2dpi, 12dpi) Results: The RNASeq analysis in SG200 identifies differences in gene expression with FB1xFB2. These differences could be the result of a unequal contribution of each nuclei to transcription. more...

Telomerase RNA subunit of Ustilago maydis

(Submitter supplied) The RNA subunit of telomerase is an essential component whose primary sequence and length are poorly conserved among eukaryotic organisms. The phytopathogen Ustilago maydis is a dimorphic fungus of the Ustilaginaceae family. We analyzed several members of the Ustilaginaceae family to computationally identify the TElomere RNA (TER) gene ter114. To confirm the identity of the TER gene, we disrupted the gene and characterized telomerase-negative mutants. more...

Comparison of extracellular vesicle and filamentous cell transcriptomes of Ustilago maydis

(Submitter supplied) The coding transcriptomes of filamentous cultures of the maize smut fungus Ustilago maydis and their extracellular vesicles (EVs) were compared. Protein-coding transcripts relatively enriched in EVs versus filament cells were identified and examined to identify potentially functional mRNA cargos of U. maydis EVs.

NRG1 as a regulator of pH adaptation in the Basidiomycota Ustilago maydis

(Submitter supplied) A homologue of the transcriptional repressor ScNrg1 described previously in the budding yeast and Candida albicans NRG1 wich plays an essential role in repressing hyphal development in both Saccharomyces cerevisiae and C. albicans, was found in Ustilago maydis. In S. cerevisiae it regulates a set of stress-responsive genes, also in Cryptococcus neoformans is involved in pathogenesis. In this work, we describe the effect of the absence of the U. more...

Ethanol induction of the yeast-to-mycelium dimorphic transition in the Basidiomycota fungus Ustilago maydis

(Submitter supplied) The sexual cycle of Ustilago maydis includes the succession of a saprophytic haploid yeast stage to a virulent hyphal dikaryotic stage, product of the mating of sexually compatible yeast cells. This dimorphic transition may be replicated in vitro by different means. Recently, it was shown that ethanol induced filamentous growth on solid medium, and we observed that this process also occurred in liquid media. more...

Transcriptome-wide view of endosomal mRNP transport [RNA-Seq]

(Submitter supplied) Spatiotemporal protein expression is mediated by active transport and local translation of mRNAs. Key factors of the transport machinery are RNA-binding proteins (RBPs) that recognise mRNAs as cargo for motor-based transport. However, a global view of transported mRNAs with cognate RBP binding sites is missing. Here, we cast a transcriptome-wide view on endosomal mRNP transport in Ustilago maydis by studying the newly identified endosomal RBP Grp1 and the key transport RBP Rrm4 in a comparative iCLIP approach. more...

Transcriptome of Ustilago maydis during tumour formation in bundle sheath and mesophyll cells of Zea mays L.

(Submitter supplied) The biotrophic fungus Ustilago maydis causes smut disease on maize (Zea mays L.), which is characterized by immense plant tumours. To establish disease and reprogram organ primordia to tumours, U. maydis deploys effector proteins in an organ-specific manner. However, the cellular contribution to leaf tumours remains unknown. We investigated leaf tumour formation on the tissue- and cell type-specific level. more...

Time series transcriptional profiling of Ustilago maydis infecting Zea mays

(Submitter supplied) The basidiomycete fungus Ustilago maydis causes smut disease in maize and has become an important model for elucidating the strategies used for host colonization by biotrophic fungi. In this study, we performed an in-depth transcriptional profiling of the plant-associated development of a cross between U. maydis FB1 and FB2 wildtype strains. The analysis of eight different stages, including the development on the leaf surface, early colonization, tumor induction and spore maturation, offers an unprecedented view of the changes in the fungal transcriptome associated with the passage through the entirely biotrophic life cycle. more...
